Combinatorial effects of microRNA to suppress the myc oncogenic pathway (miRNA data)

ABSTRACT: microRNA importance in the myc pathway in Burkitt lymphoma. MicroRNA data from the Agilent human miRNA platform. 12 Burkitt lymphomas were compared with 14 non-tumoral controls (lymph nodes, tonsils and CD19+CD10+ sorted cells). Significantly deregulated miRNAs were computed using the Significant Analysis of Microarray (SAM) analysis from the TM4 pathway and the limma package.

Project description:Many mammalian transcripts contain target sites for multiple miRNAs, although it is not clear to what extent miRNAs may coordinately regulate single genes. We have mapped the interactions between down-regulated miRNAs and overexpressed target protein-coding genes in murine and human lymphomas. Myc, one of the hallmark oncogenes in these lymphomas, stands out as the up-regulated gene with the highest number of genetic interactions with down-regulated miRNAs in mouse lymphomas. The regulation of Myc by several of these miRNAs is confirmed by cellular and reporter assays. The same approach identifies MYC and multiple Myc targets as a preferential target of down-regulated miRNAs in human Burkitt lymphoma, a pathology characterized by translocated MYC oncogenes. These results indicate that several miRNAs must be coordinately down-regulated to enhance critical oncogenes, such as Myc. Some of these Myc-targeting miRNAs are repressed by Myc, suggesting that these tumors are a consequence of the unbalanced activity of Myc versus miRNAs. Project description:The effects of Gata4 inhibition on global gene expression in TM4 Sertoli cells was analyzed. Using microarray analysis we identified a set of genes that are downregulated or upregulated following siRNA-mediated silencing of Gata4 in the murine Sertoli cell line TM4. Among the downregulated genes were regulators of blood-testtis barrier function and lactate metabolism. TM4 cells were transfected with non-targeting (NT) siRNA and Gata4 siRNA respectively. 72h post transfection cells were lysed and RNA was extracted.
