Colour analysis in Fenneropenaeus merguiensis

ABSTRACT: Differential gene expression analysis of three F. merguiensis colour groups (dark, light, albino), using custom microarrays. A total of 12 light, 12 dark and 9 albino prawn samples were analysed, using total RNA extracted from cuticle tissue. 3 biological replicates were pooled onto each of the 4 arrays for the light and dark samples; and for the albino samples, groups of 2, 2, 2 and 3 biological replicates were pooled for the 4 arrays. Single colour analysis was carried out, using the Cy3 dye.

Miyazawa2010 - changing turing patterns by spatial parameter sweep

Project description:Biologists have long been fascinated by the amazing diversity of animal colour patterns. Despite much interest, the underlying evolutionary and developmental mechanisms contributing to their rich variety remain largely unknown, especially the vivid and complex colour patterns seen in vertebrates. The model [1] shows, that complex and camouflaged animal markings can be formed by the ‘blending’ of simple colour patterns. A mathematical model predicts that crossing between animals having inverted spot patterns (for example, ‘light spots on a dark background’ and ‘ dark spots on a light background’ ) will necessarily result in hybrid offspring that have camouflaged labyrinthine patterns as ‘ blended ’ intermediate phenotypes. In [1] the authors confirmed the broad applicability of the model prediction by empirical examination of natural and artificial hybrids of salmonid fish. The results suggest an unexplored evolutionary process by means of ‘pattern blending’, as one of the possible mechanisms underlying colour pattern diversity and hybrid speciation. The model file is in MorpheusML format and can be opened in the free, open-source multicellular modeling software Morpheus (https://morpheus.gitlab.io) to simulate the time course (movie) of the spatio-temporal dynamics. It resembles figure 3a from [1], where a 2D parameter space shows the transition from black to white spots through intermediate labyrinthine patterns. 1. Impact of albino chemical on Arabidopsis seedlings

Project description:Analysis of wild type genome expression profile altered by albino chemicals. Results uncovered a several groups of genes that were significant responsive to these chemicals which may contribute to the albino phenotype. WT (Col-0) sown on 0.5x MS agar medium and stratified in the dark at 4 Â°C for 3 days and then germinated in the light condition (16 h light/8 h dark) for 5 days at 22 Â°C, and then transfer to the 0.5x MS solution one day for adapt the treatment condition. In the DAG 6 (day after germination), final concentration 50 Î¼M chemicals (7606596 and 5929745) were supplemented into the medium for chemical treatment. Approximately 2,000 seedlings were used to collect tissues before the chemicals treatment as the time zero samples. Seedlings were collected after the chemical treatment for 3 hours and 12 hours. Total RNA extracted from the collected samples was applied in each microarray experiment with Arabidopsis ATH1 GeneChip (Affymetrix). Qiagen RNAeasy mini kit (Qiagen) was used for RNA preparation. cDNA labeling, array hybridization, and scanning followed by standard Affymetrix expression array protocol. Two independent sets of microarray analyses were performed in this assay.

Gene expression profiling in three inbred lines of maize affected by chilling treatment

Project description:We investigated chilling response of seedlings of three inbred maize lines: chilling tolerant S68911, chilling-sensitive S160 and moderate chilling-sensitive S50676. Kernels were germinated in wet sand in darkness at 25C. Seedlings were transferred to growth chamber (photoperiod 14/10h, temperature 24, light 250 umol quanta x m-2 x s-1), grown till the third-leaf stage and used in experiment. Chilling treatment started at the start of the dark period and lasted 38h (10h dark, 14h light,10h dark, 4h light). Growth conditions was as previously described but temperature was set to 14 (light/dark). At the same time control plants were grown as previously described. There were three biological replications of hybridization scheme.
