Gene expression in 16 dpp Tex19.1-/- mouse testes

ABSTRACT: Microarray experiment to identify changes in gene expression in 16 day post partum prepubertal Tex19.1-/- mouse testes. Tex19.1 is expressed in germ cells in testes. Tex19.1-/- mice have spermatogenic defects and errors in progression through meiosis. Data provides insight into the changes in gene expression in developing testes at the time when meiotic defects first start to become apparent. Testes from four Tex19.1-/- animals (ko) and four Tex19.1+/- littermate controls (het) are included in the analysis.

Project description:Deleted in Azoospermia Associated Protein 1 (DAZAP1) is a ubiquitous RNA-binding protein that is highly expressed in the testis. It is a component of the hnRNP particles and shuttles between the nucleus and the cytoplasm. Mice expressing the DAZAP1-Fn mutant protein manifest both growth retardation and spermatogenic arrest before meiosis I. To elucidate the biological function(s) of DAZAP1 and to search for its natural RNA substrates, we compared the expression profiles of wild-type and Dazap1 mutant testes by cDNA microarrays. We used wild-type and Dazap1 mutant mouse testes. Each genotype has three replicates. 6 total samples were analyzed.

Project description:Activation of the Heat Shock Factor 1 (HSF1) in spermatogenic cells leads to apoptosis and infertility of males. To elucidate a mechanisms of apoptosis induced by HSF1 we generated transgenic mice expressing mutated, constitutively active human HSF1 (with a deletion of amino acids 221–315) specifically in spermatocytes (under control of the rat Hspa2 promoter). We carried out genome-wide transcriptional analyses in testes of control, wild-type, and transgenic males. Genes that changed the expression due to activation of HSF1 have been identified.

DICER-interacting partners in juvenile mouse testes

Project description:DICER has a well-characterized role in the processing of microRNAs (miRNAs) and small interfering RNAs (siRNA) that are important for post-transcriptional gene regulation. Emerging evidence suggests that DICER also has several non-canonical functions beyond miRNA/siRNA biogenesis, for example in transcriptional gene silencing at the chromatin level, as well as in RNA degradation and maintenance of genomic integrity. We have shown that the function of DICER in germ cells is essential for normal spermatogenesis; male mice lacking DICER in postnatal male germ cells are infertile due to severe defects in haploid differentiation. To better understand the function of DICER in male germ cells, we immunoprecipitated DICER from juvenile mouse testes and performed mass spectrometric analysis to identify DICER-interacting proteins.
