ABSTRACT: Maternal and fetal monocytes and tissue macrophages (decidual macrophages, Hofbauer cells) at the feto-maternal interface have different methylome. Paired and balanced design. We compared maternal blood monocytes (MB) vs. cord blood monocytes (CB), maternal blood monocytes (MB) vs. decidual macrophages (Deci), cord blood monocytes (CB) vs placental macrophages (villi) and decidual macrophages (Deci) vs. placental macrophages (villi).

Single-cell sequencing and second-generation RNA sequencing-based screening of macrophage-associated marker genes for recurrent miscarriage diagnosis

Project description:We profiled the transcriptomes of about 110000 single cells from 16 human first-trimester decidual and villi samples from 5 normal samples and 3 recurrent miscarriage (RM) samples. Each decidual and villi samples was collected from the same patient. The cellular composition revealed five major subsets of immune cells including NK, T cell, macrophage, monocytes and B cell in the maternal fetal interface. We used the marker gene of macrophages to establish the diagnosis and treatment model of unexplained recurrent abortion, in order to explore the relationship between immune cells and unexplained recurrent abortion.

Transcription profiling of human decidual macrophages

Project description:Uterine macrophages are thought to play an important regulatory role at the maternal-fetal interface. We report here a unique gene expression pattern intrinsic of first trimester decidual monocytes/macrophages, but not of their blood counterparts. The micro-array data comprises approximately 14,000 genes. Some of the key findings were confirmed by real time PCR or secreted protein measurements. A large number of regulated genes were found to be functionally related to immunomodulation and tissue remodelling, corroborating polarization patterns of differentiated macrophages of M2 phenotype. These include M2 markers such as CCL-18, CD209, insulin-like growth factor (IGF)-1, mannose receptor c type (MRC)-1 and fibronectin-1. Further, the selective up-regulation of triggering receptor expressed on myeloid cells (TREM)-2, alpha-2-macroglobulin (A2M) and prostaglandin D2 synthase (PGDS) provides new insights into the regulatory function of decidual macrophages in pregnancy. In addition, a large number of regulated genes in the micro-array analysis were related to cell cycle regulation. Taken together, molecular characterization of decidual macrophages presents a unique transcriptional profile replete with important components for fetal immunoprotection. Human placental exosomes induce maternal systemic immune tolerance in a monocyte-dependent manner

Project description:Maternal immune tolerance toward to the semi-allograft fetus is a prerequisite condition for successful pregnancy. However, the role of maternal monocytes in the induction of systemic immune tolerance is poorly understood. Here we report that placenta facilitates maternal immune tolerance by extruding exosomes. Maternal monocytes were transformed into an immunosuppressive phenotype after taking up exosomes. Mechanistically, PD-L1 was significantly increased in pEXO-educated monocytes via miRNA-29a-3p/PTEN signaling pathway. In addition, pEXO-treated monocytes could increase Treg pool in maternal blood through a direct cell-cell interaction. Moreover, Th1 cytokines, IFNγ and TNF-α, in monocyte-T cell co-culture system were significantly decreased. Together, our results indicated that maternal monocyte is indispensable components in systemic immune tolerance establishment.

Single-cell immunophenotyping of the fetal immune response to maternal SARS-CoV-2 infection

Project description:During the COVID-19 pandemic, thousands of pregnant women have been infected with SARS-CoV-2 worldwide. The short- and long-term implications of maternal SARS-CoV-2 infection on fetal and childhood well-being are unknown. To characterize the fetal immune response to maternal SARS-CoV-2 infection, we performed single-cell RNA sequencing and T cell receptor sequencing on cord blood mononuclear cells from infants born to mothers infected with SARS-CoV-2 in the third trimester. We identified widespread gene expression changes in cord blood leukocytes, including upregulation of interferon-stimulated genes (ISG) and of HLA genes in CD14+ monocytes; decreased activation of CD16+ monocytes; activation of plasmacytoid dendritic cells; and activation and exhaustion of NK cells and T CD8+ cells in the cord blood of infants born to SARS-CoV-2+ mothers. Lastly, we observed fetal TCR repertoire clonal expansion in cord blood T cells from pregnancies complicated by maternal SARS-CoV-2 infection. Our results suggest that even in the absence of vertical transmission, SARS-CoV-2 maternal infection in the third trimester modulates the fetal immune system.

Project description:Maternal body size, nutrition, and hyperglycemia contribute to neonatal body size and composition. There is little information on maternal-fetal transmission of messages which influence fetal growth. We analyzed adipocyte-derived small extracellular vesicular (ADsEV) microRNAs in maternal and cord blood to explore their adipogenic potential. Differential expression (DE) of ADsEV miRNAs in adipose vs. lean neonates was studied before and after adjustment for maternal gestational diabetes mellitus (GDM), adiposity, and vitamin B12-folate status.
