Transcription profiling of rats (SHR) on normal and high salt plus +fat diets, at 2 vs. 8 weeks

ABSTRACT: Supporting microarray data for manuscript entitled "OSTEOPONTIN AND PAI-1 EXPRESSION IN MALIGNANT HYPERTENSION: SUPPRESSION BY p38 MAPK INHIBITORS" submitted to the HYPERTENSION journal. Experiment Overall Design: Male spontaneously hypertensive stroke-prone rats (SHR-SP) were obtained from Charles River (Raleigh, NC). At 11 weeks of age, the SHR-SP were randomized into 2 groups, and fed either powdered chow diet (Purina 5001) with water ad lib; or a high-salt/high-fat diet consisting of 1% NaCl in the drinking water and 24.5% fat in the chow (from Harlan TekLad, Madison, Wisconsin). 6 replicate animals per diet per time point.

Transcription profiling of liver and white adipose tissue from mice fed standard or test diets with or without monosodium glutamate in their water

Project description:C57BL/6J mice were from The Jackson Laboratory (Maine, USA) and were housed/caged and fed a standard chow diet until 6 weeks of age whereupon they were placed in one of 4 different dietary regimens for a period of 3 weeks prior to mating. Mice were housed under pathogen-free conditions of 21± 1°C, 40 to 60% humidity, and a 12 h-12 h light/dark cycle. The 4 diet regimens used in this study were: [1] ad lib Standard Chow (Control diet) with ad lib drinking water. [2] Ad lib Standard Chow, with ad lib drinking water containing 0.64 mg/ml (97 mg/Kg body weight) monosodium glutamate (MSG diet). [3] Ad lib Test Diet Purina 5001 with 20% 55-HFCS (5B4K, Purina, USA), and ad lib drinking water (HFCS diet). [4] Ad lib Purina 5001 with 20% 55-HFCS, and ad lib drinking water containing 0.64 mg/ml (97 mg/K body weight) monosodium glutamate (HFCS+MSG diet). See Table 1 for diet composition. Following mating, the 4 groups of dams were maintained on their respective diets throughout the gestation and nursing period. Male offspring used in these experiments were weighed, then weaned onto the same diets and maintained in this regimen until they reached either 16 or 32 weeks of age. Total RNA was prepared from the liver and visceral white adipose tissue (WAT) taken from 16-week-old mice in the 4 different diet groups using Qiagen RNeasy Kit (Qiagen USA) according to the manufacturers instructions and stored at -80 o C.<br>

microRNA expression profiles in myocardium of diet-induced obesity rat

Project description:Diet induced obesity in rat was associated with myocardial dysfunction, hypertension and fibrosis. This study aimed to explore microRNA expression profiles in diet obesity-induced rat myocardium. Wistar rats were feed normal chow or high-fat diet for 20 weeks. After that, cardiac function was evaluated by echocardiography. Left ventricular myocardium was harvest to assess the extent of hypertension and fibrosis, meanwile, the left ventricular microRNA expression was analyzed using Agilent Rat miRNA microarray. Significant cardiac dysfunction, hypertension and fibrosis were found in diet-induced obesity rats as compared with normal diet rats. rno-miR-141-3p and rno-miR-144-3p were also significantly increased in myocardium of diet-induced obesity rat. These findings suggest that specific miRNA differences may contribute to the alteration in cardiac function, hypertension and fibrosis which responses to diet-induced obesity.

Transcription profiling of mouse lung from animals exposed to arsenic

Project description:Exposure to high levels of arsenic in drinking water is associated with several types of cancers including lung, bladder and skin, as well as vascular disease and diabetes. Drinking water standards are based primarily on epidemiology and extrapolation from higher dose experiments, rather than measurements of phenotypic changes associated with chronic exposure to levels of arsenic similar to the current standard of 10ppb, and little is known about the difference between arsenic in food as opposed to arsenic in water. Measurement of phenotypic changes at low doses may be confounded by the effect of laboratory diet, in part because of trace amounts of arsenic in standard laboratory chows, but also because of broad metabolic changes in response to the chow itself. Finally, this series contrasts 8hr, 1mg/kg injected arsenic with the various chronic exposures, and also contrasts the acute effects of arsenic, dexamethasone or their combination. Male C57BL/6 mice were fed on two commercially available laboratory diets (LRD-5001 and AIN-76A) were chronically exposed, through drinking water or food, to environmentally relevant concentrations of sodium arsenite, or acutely exposed to dexamethasone. Experiment Overall Design: Male C57BL/6 mice, fed on two commercially available laboratory diets (LRD-5001 and AIN-76A), were chronically exposed through drinking water or food, to environmentally relevant concentrations of sodium arsenite. Another group animals, fed on the AIN 76A diet, was IP injected with dexamethasone (1 mg/kg), sodium arsenite (1mg/kg), both dexamethosone and arsenite, or saline alone.
