Transcription profiling of 5- and 10-week bitransgenic mouse mammary glands (LH x MMTV-neu) to identify genes that may facilitate early steps of ErbB2/Neu-mediated mammary tumorigenesis.


ABSTRACT: To identify genes that may facilitate early steps of ErbB2/Neu-mediated mammary tumorigenesis, we performed comparative microarray analysis of 5- and 10-week bitransgenic mammary glands (LHxMMTV-neu) in triplicate. Experiment Overall Design: We analyzed 3 pooled RNA samples that represented 3 animals for each time point so that a total of 9 bitransgenic mice were analyzed per time point.

Epidemiological studies indicate that parity enhances HER2/ErbB2/Neu-induced breast tumorigenesis. Furthermore, recent studies using multiparous, ErbB2/Neu-overexpressing mouse mammary tumor virus (MMTV-Neu) mice have shown that parity induces a population of cells that are targeted for ErbB2/Neu-induced transformation.
