Development of gene expression signatures for mesenchymal stem cells from neuroblastoma patients

ABSTRACT: To obtain genetic evidence about the interaction between MSCs and metastatic NB cancer cells in bone marrow (BM), we isolated bone marrow-derived MSCs from children with NB and compared their global expression patterns with MSCs obtained from normal pediatric donors using the Agilent 44K microarrays. A total of eight different donors (four NB patients and four healthy donors) were analyzed and three independent microarrays hybridizations were performed for each donor.

Project description:Administration of mesenchymal stem cells (MSCs) has the potential to ameliorate degenerative disorders and to repair damaged tissues. The homing of transplanted MSCs to injured sites is a critical property of engraftment. Our aim was to identify microRNAs involved in controlling MSC proliferation and migration. MSCs can be isolated from bone marrow and umbilical cord Wharton's jelly (BM-MSCs and WJ-MSCs, respectively), and WJ-MSCs show poorer motility yet have a better amplification rate compared to BM-MSCs. One human BM-MSC (pooled sample of 4 independent donors), one human WJ-MSC (pooled sample of 3 independent donors), and differentiated osteocytes and adipocytes derived from BM-MSCs after 2 weeks induction.

Gene expression profiling of primary human bone marrow mesenchymal stromal cells (MSCs) cultured as 2D monolayers and 3D cellular aggregates (spheroids)

Project description:Human bone marrow mesenchymal stromal cells (MSCs) are conventionally cultured as adherent monolayers on tissue culture plastic. MSCs can also be cultured as 3D cell aggregates (spheroids). Optimised 3D conditions (60,000 MSCs cultured as a spheroid for 5 days) inhibited MSC proliferation and induced cell shrinkage in the absence of cell death. Primary human MSCs isolated from 2 donors were cultured under both monolayer (2D MSCs) and optimised 3D (3D MSCs) conditions. High quality RNA was isolated from all samples, and global gene expression analysis was performed in duplicate (using Agilent SurePrint G3 Human Gene Expression 8x60K v2 Microarrays) to identify gene expression changes in 3D compared to 2D MSC cultures.

Gene expression patterns of mesenchymal stem cells from bone marrow and Wharton's jelly of umbilical cord

Project description:Mesenchymal stem cells (MSCs) are promising tools for the treatment of diseases such as infarcted myocardia and strokes because of their ability to promote endogenous angiogenesis and neurogenesis via a variety of secreted factors. MSCs found in the Wharton's jelly of the human umbilical cord are easily obtained and are capable of transplantation without rejection. We isolated MSCs from WhartonM-bM-^@M-^Ys jelly and bone marrow (WJ-MSCs and BM-MSCs, respectively) and compared their transcriptional profiles. In this study, BM-MSCs and WJ-MSCs were isolated from 4 and 3 independent donors, respectively.

Transcriptomic signature induced in bone marrow Mesenchymal Stromal Cells (MSC) after interaction with malignant multiple myeloma cells: implications in myeloma progression.

Project description:Mesenchymal stromal cells (MSCs) derived from the BM of healthy donors (dMSCs) and myeloma patients (pMSCs) were co-cultured with the model myeloma cell line - MM.1S -, and the gene expression profile of MSCs induced by this interaction was analyzed using high density oligonucleotide microarrays. Deregulated genes in co-culture common to both d/pMSCs revealed functional involvement in tumor microenvironment cross-talk, myeloma growth induction and drug resistance, angiogenesis and signals for osteoclast activation and inhibition of osteoblasts. Additional genes induced by co-culture were exclusively deregulated in pMSCs and were predominantly associated to RNA processing, the ubiquitine-proteasome pathway, regulation of cell cycle and Wnt signaling. Primary MSCs from bone marrow (BM) samples of healthy donors (n=8) and multiple myeloma (MM) patients (n=13) were generated as described by Garayoa et al. 2009 (PMID:19357701). Ficoll-Paque density gradient separation medium was used to isolate mononuclear cells from BM aspirates. MSCs were selected by adherence to plastic and subsequently expanded until passage 2 when they were used for co-culture. For the MSC-MM.1S co-cultures we used a 6-well format transwell system with 1 M-BM-5m pore size membrane. Briefly, 2 X 10^4 MSCs were first cultured attached to the lower side of the membrane, and when reached M-bM-^IM-^H 85% confluence 1 X 10^6 MM.1S cells were seeded on the upper side. After 24 h co-culture in RPMI 1640 medium supplemented with 10% FBS and antibiotics, MSCs were recovered by trypsinization, washed once in PBS and stored at -80M-BM-0C in RLT buffer (Qiagen) until RNA extraction. Absence of MM.1S cells in the recovered MSC population was assessed in selected samples by flow cytometry analysis.

Project description:This experiment investigates differences in miRNA expression between different populations of human mesenchymal stem cells (MSCs), which are a potential treatment for multiple sclerosis (MS). Previously it has been reported that MSCs derived from human olfactory mucosa (OM-MSCs) may be a better candidate for the treatment of MS than MSCs derived from bone marrow (BM-MSCs), due to a better ability to promote CNS myelination in vitro. In this study, miRNA profiling of OM-MSCs and BM-MSCs was undertaken to investigate the differences between these two cell types in relation to their prospective therapeutic use.

In this study, hBM-MSCs were isolated from residual human bone marrow (hBM) material and expanded to clinically relevant numbers at passage 3-4. The cells had normal morphology, proliferation rate, immunophenotype and karyotype. Despite the chromosomal stability, after additional three passages (P6-P7) hBM-MSCs entered senescence state, confirmed by SA-Î²-galactosidase staining and paralleling the slower proliferation, altered morphology and imunophenotype. qRT-PCR array profiling revealed 3 out of 86 genes significantly (p <0.05) differentially (â¥2 fold) expressed in the late passage cells compared to the early passage cells. qPCR gene expression profiling. hBM-MSCs from 3 (1 male and 2 females) adult donors (age 30Â±7.21 years) were used. Each sample was tested in technical duplicate. 6 samples of early passage (P3-P4) hBM-MSCs were grouped to CONTROL group and 6 samples of late passage (P6-P7) hBM-MSCs were grouped to TEST group. The data were analyzed using web-based RT2 Profiler PCR Array Data Analysis v3.5 software (Qiagen)
