Differentially expressed genes in recurrence of squamous cell carcinoma tumors of the oral cavity in a mouse model

ABSTRACT: We applied global gene expression with samples derived from a recently established mouse model for oral cancer recurrences and identified a list of genes with differential expression between primary and recurrent tumors. 5 independent primary tumors and respectivce recurrences were analyzed. Two replicates of the original cell line used for the generation of all tumors was also added to the study.

Project description:New targets and drugs for treatment of breast cancer are needed. We tested the effects of the TKI258 (Novartis) FGFR-inhibitor on tumor formation in mice. 4T1 mouse cell line, which had been derived from BALB/c mice, was injected into tail veins of BALB/c mice (syngraft model). Seven days later, mice were treated with 0, 15 or 40 milligram per kilogram (dose) TKI258 inhibitor (compound). Tumors were resected after different time points (time) and effects of drug treatment on global gene expression was monitored.

Gene expression profiling of primary breast carcinomas from patients at high risk for local recurrence

Project description:Purpose: Several risk factors for local recurrence of breast cancer after breast conserving therapy (BCT) have been identified. The identification of additional risk factors would be very useful in guiding optimal therapy and also improve understanding of the mechanisms underlying local recurrence. We used cDNA microarray analysis to identify gene expression profiles associated with local recurrence. Experimental Design: Using 18K cDNA microarrays, gene expression profiles were obtained from 50 patients who underwent BCT. Of these 50 patients 19 developed a local recurrence; the remaining 31 patients were selected as controls as they were free of local recurrence at least 11 years after treatment. For 9/19 patients also the local recurrence was available for gene expression profiling. Unsupervised and supervised methods of classification were used to separate patients in groups corresponding to disease outcome and to study the overall gene expression pattern of primary tumors and their recurrences. Results: Hierarchical clustering of patients did not show any grouping reflecting local recurrence status. Supervised analysis revealed no significant set of genes that was able to distinguish recurring tumors from non-recurring tumors. Paired-data analysis of primary tumors and local recurrences showed a remarkable similarity in gene expression profile between primary tumors and their recurrences. Conclusions: No significant differences in gene expression between primary breast cancer tumors in patients with or without local recurrence after breast conserving therapy were identified. Furthermore, analyses of primary tumors and local recurrences show a preservation of the overall gene expression pattern in the local recurrence, even after radiotherapy. Combined external beam radiotherapy (photon or carbon) and 131I-MIP-1145 (benzamide) endoradiotherapy in syngeneic B16F10 mouse tumor model

Project description:Subcutaneous B16F10 tumors in the right hind limb of C57BL/6 mice were treated with 5 daily fractions of external beam radiotherapy (8 Gy photon or 5 Gy carbon per fraction), a single fraction of 13.3 MBq 131Iodine-labelled benzamide-derivative MIP-1145 intravenously or a combination of the two. Untreated tumors served as controls. Mice were sacrificed and tumor tissue collected for expression profiling 5 days after endoradiotherapy of 1 week after the last fraction of external beam radiotherapy.

Phospho β-catenin Serine552 Predicts Recurrence of Non-Functioning Pituitary Tumors After Surgery

Project description:NFPTs are tumors without a clinical evidence of hormone hypersecretion. Due to their lack of clinically detectable hormonal activity, they tend to present with mass effects. The molecular pathogenesis of NFPT is not well understood. Treatment of these non-functioning tumors is either surgery or radiotherapy. When recurrent, they are associated with severe morbidity. Characterization of phosphorylation status is critical to elucidate signaling pathways leading to tumorigenesis. Therefore, identifying phosphorylated proteins and their sites of phosphorylation will provide an important clue to the mechanism of NFPT pathogenesis, they can be used as predictor of aggressive behavior as well as activated kinases may be specifically targeted using small molecule inhibitors. Till date no study has undertaken global analysis of phosphoproteins in NFPTs. In this study, we designed an integrative approach involving identification of aberrant phosphorylation events in NFPTs using quantitative (Tandem mass tag-based) mass spectrometry based phosphoproteomic profiling followed by in-silico analysis and validation of candidate phosphoproteins in large cohort of NFPTs.

Project description:Nu61, a radiation-resistant human tumor xenograft, was selected from a parental radiosensitive tumor SCC-61 by eight serial cycles of passage in athymic nude mice and in vivo irradiation. Obtained tumors were profiled using Affymetrix U133A arrays. Most abundant gene pattern associated with radioresistant phenotype was presented by IFN-inducible, Stat1-dependent pathway Experiment Overall Design: Obtained tumors were established as xenografts in nude mice. RNA were purified from each xenograft, normalized by concentration and used for hybridization with each array. FGFR2 Signaling Underlies p63 Oncogenic Function in Squamous Cell Carcinoma

Project description:This study was designed to examine the requirement for the p63 transcription factor in Squamous Cell Carcinoma (SCC) tumor maintenance in an in vivo murine system. A tamoxifen-inducible Keratin 14-driven Cre recombinase transgene was used to conditionally excise p63 in advanced murine SCC tumors. These data show the context-dependent regulation of p63 target genes in cancer. Total RNA from murine Squamous Cell Carcinoma tumors was examined 1-3 days following genomic excision of TP63 in Keratin 14-expressing tumor cells.
