ABSTRACT: The microarray analysis was used for comparing the gene expression profiles between MEF, MEF treated with n-Butylenephthalide (BP) 10 and 40ug/ml. The total RNA extracted from MEF, MEF treated with n-Butylenephthalide (BP) 10 and 40ug/ml was used for the microarray analysis to compare the gene expression profiles.

Project description:Gene expression of Retinoblastoma deficient mouse embryonic fibroblasts (Rb-/- MEF, ME3) and wildtype mouse embryonic fibroblasts (Rb+/+ MEF,MEFA) during adipocyte differentiation were studied by cDNA microarray analysis. 2-day postconfluent cells (day 0) were treated with a hormonal cocktail including Rosiglitzone for differentiation into adipocytes. Gene expression at day 1, day 3, and day 8 after hormonal induction were compared to that before induction (day 0).

Project description:In a pilot experiment to reprogramme MEF into endoderm, we infected MEF with the YamanakaM-BM-4s factors (O: Oct4, K: Klf4, S: Sox2, M:Myc), FoxA2 (F) and Gata4 (G). Global gene expression of isolated clones was performed. After infection of MEF with the YamanakaM-BM-4s factor (OKS/M), FoxA2 and Gata4, the cells were grown until colonies could be isolated and expanded independently.
