Expression data from st 12-13 embryos from 2xbcd and 6xbcd mothers

ABSTRACT: Embryo from 6xbcd mother shows expansion of prospective head region. In this embryo, excessive cell death can be observed. This array expreriment is for identifying differentially expressed genes in the 2xbcd and 6xbcd conditions at stage 12-13 We crossed TT16 males with TT16 females for the 2xbcd condition and bcd+t8 females for 6xbcd. In both crosses, eggs were collected in 90 minutes and incubated for 8 hours at 25°. After excluding unfertilized eggs under stereomicroscope, we extracted RNA from more than 100 eggs and hybridized by GeneChip Drosophila Genome 2.0 arrays (Affymetrix) for hybridization. We carried out paired (2xbcd vs. 6xbcd) microarray experiments in three replicates using independent RNA samples.

Project description:A key feature of Nasonia are their form of sex determination, called haplodiploidy. Females are diploid and develop from fertilized eggs, whereas males are haploid and develop parthenogenetically from unfertilized eggs. This form of sex determination is exploited by this experiment, by knowing the sex of the organisms from the earliest stages of development, so to trace the transcriptomes of sexual development of an insect. We conducted three replicates each using RNA from independent biological extractions of male and female early embryo (0-10 hrs), late embryo (18-30 hrs), 1st instar larvae, and pupae. Additional experiments were performed comparing transcription in adult males, adult females, testis and the female reproductive tract.

Broodstock - transcription profiling of maternal RNA in Atlantic cod eggs and embryos after female broodstock stress

Project description:A stable supply of viable eggs and embryos is crucial for successful farming of Atlantic cod. Broodstock stress can have negative effects, but little is known about the molecular mechanisms that cause abnormal development. Maternally transferred mRNAs have been shown to be essential for normal development, and stress may therefore influence their expression and the subsequent embryonic development. We investigated if mimicked stress in prespawning Atlantic cod females affects mRNA concentrations in eggs and embryos, and if this can be linked to egg/embryo viability, and if we could identify potential molecular markers for stress and egg/embryo viability. Consequently, 3 weeks prior to expected peak spawning, 20 females were intraperitoneally implanted with either cortisol-containing or cortisol-free (sham) osmotic pumps. At peak spawning all individuals were stripped and eggs were fertilized and incubated until hatching. Samples were collected from unfertilized eggs and embryos for gene expression profiling.
