ABSTRACT: Analysis of gene expression in FACs-sorted pre-B cells derived from Fnip1-null and normal wildtype littermate mice. 3 arrays from 3 different mice per genotype were utilized to generate the expression data (mouse WG-6 v.2.0 beadchip (Illumina Inc, San Diego CA)) and statistics. P-values are shown. Total RNAs were isolated from sorted B220+CD43+ cell from Fnip1-/- and WT mice (n=3)

BTG1 and BTG2 Regulate B Linege Commitment by Modulation of EBF1 Activity

Project description:The human BTG1 and BTG2 genes are targeted by genetic alterations in hematological malignancies. In this study we examined the unique and overlapping functions of Btg1 and Btg2 in mouse B cell development. Mice deficient for Btg1 and Btg2 displayed the strongest phenotype with a decrease in B cell numbers in bone marrow due to impaired B cell commitment and immature B cell differentiation. To determine the underlying mechanism, microarray expression analyis was performed on early (B220+ CD43+) and late (B220+ CD43-) B cells of WT and Btg1;Btg2 dKO mice. Total RNA obtained from sorted B220+ CD43+ ( Hardy fractions ABC) and B220+ CD43- (Hardy fractions DEF) BM cells of WT and Btg1-/-;Btg2-/- mice. In total 4 samples were analyzed and no replicates were included.

Project description:We used the NanoString mouse nCounter miRNA expression platform to compare the miRNA expression profiles of pro-B cells sorted from fetal liver and adult bone marrow in order to gain insight into changes in miRNA expression during B cell ontogeny. Fetal liver and adult bone marrow cells were harvested and FACS sorted for pro-B cells (B220+CD43+IgM-veCD19+CD24+). Expression data from murine bone marrow-resident plasma cells and spleen mature follicular B cells

Project description:CD138+ B220- plasma cells were sorted from bone marrow and B220+ CD23+ mature follicular B cells were sorted from the spleens. Plasma cells were sorted from C57BL/6 mice 7 days after boosting with antigen, with mice first primed with an i.p. injection of KLH/IFA followed by boost at day 21 with KLH/PBS i.p. Mature B cells were sorted from antigen-naïve C57BL/6 mice. We compared expression profiles of plasma cells and mature B cells to identify differentially expressed transcripts.
