Comparison of Control and Tim-Fbl gene expression profiles

ABSTRACT: In order to identify genes affected in our model of PANK defficiency we profiled fly heads of female flies control or expressing fumble RNAi by the use of the tim-gal4 driver Female fly heads of the indicated strains were collected at 2 different circadian timepoints, RNA extracted, labeled and hybridized to Affymetrix oligonucleotide microarrays.

Fly heads from GAL4 heterozygotes were used as controls. The hits from FLAG-tagged twenty-four (TYF) proteomics were also excluded from the list to increase TANGO10-specificity. Proteins listed are those present in at least one elav-GAL4 sample and at least one tim-GAL4 sample but no negative controls.

Project description:Control (+/cbtE1-UAS-cbt RNAi) or cabut RNAi flies (Tim-gal4, UAS-cbt RNAi) were starved for 16 hours and then exposed to food containing different concentrations of sucrose: 0, 25, 50 and 100 % for 18 hours. Fly heads were collected, RNA was extracted and RNA-seq libraries were prepared as previously described (Engreitz et al., 2013) For each sucrose concentration, two samples of cabut RNAi flies and one sample of control flies were sequenced.
