Expression time-series from Streptomyces coelicolor germination

ABSTRACT: Transcriptome time-courses during the S. coelicolor germination were used to reveal processes that underlie the gene expression at the transcriptional level within transition from dormancy to vegetative growth. Samples from 13 time points at the initial 5.5 h (30 min time intervals) of germination. 2-3 biological (and technical) replicates for each time point. Altogether 37 samples.

Analysis of germination and outgrowth of sorbic acid-stressed Bacillus cereus ATCC 14579 spores.

Project description:Sorbic acid (SA) is widely used as a preservative, but the effect of SA on spore germination and outgrowth has gained limited attention up to now. Therefore, the effect of sorbic acid on germination of spores of B. cereus strain ATCC 14579 was analyzed both at phenotype and transcriptome level. Spore germination and outgrowth was assessed at pH 5.5 without and with 0.75, 1.5 and 3.0mM (final concentrations) undissociated sorbic acid (HSA). This resulted in distinct HSA concentration-dependent phenotypes, varying from delays in germination and outgrowth to complete blockage of germination at 3.0mM HSA. The phenotypes reflecting different stages in the germination process could be confirmed using flow cytometry and could be recognized at transcriptome level by distinct expression profiles. In the absence and presence of 0.75 and 1.5mM HSA, similar cellular ATP levels were found up to the initial stage of outgrowth, suggesting that HSA-induced inhibition of outgrowth is not caused by depletion of ATP. Transcriptome analysis revealed the presence of a limited number of transcripts in dormant spores, outgrowth related expression, and genes specifically associated with sorbic acid stress, including alterations in cell envelope and multi-drug resistance. The potential role of these HSA-stress associated genes in spore outgrowth is discussed. Per concentration of undissociated sorbic acid (0, 0.75, and 1.5mM) four exposure times (10, 30, 60, and 120 minutes) were each compared with dormant spores (i.e., t0). Cytosolic copper concentration modulates spore germination and secondary metabolism in Streptomyces coelicolor

Project description:We studied the influence of copper in physiological and morphological differentiation of Streptomyces coelicolor. We demonstrate differences in phenotype (germination, growth rate, antibiotic production) and genetic expression between a strain mutated at copper chaperone CopZ (SCO2730::Tn5062), the wild-type strain and a wild-type strain sporulated in a media with 80µM CuSO4. These differences are correlated with the cytosolic copper. Our results demonstrate a pleiotropic effect of copper modulating S. coelicolor development.

Project description:The process of Saccharomyces cerevisiae spore germination includes breakage of dormancy, morphological changes and resumption of vegetative growth. We have determined the global transcriptional response during the first two hours of spore germination in response to rich growth medium and glucose alone, and identified possible transcription factors regulating the different transcriptional programs. Saccharomyces cerevisiae Y55 spores subjected to YPD and glucose 2%. Samples are taken in triplicates (except for glucose 0 min were duplicates were taken) in time course series after glucose and nutrient addition. Total of 18 samples. RNA from dormant spores was used as reference RNA for all microarrays.

Proteomic analysis of Streptomyces coelicolor vesicles

Project description:Streptomyces coelicolor is a model organism for the study of Streptomyces, a genus of Gram-positive bacteria that undergoes a complex life cycle and produces an enormous repertoire of bioactive metabolites and extracellular enzymes. This study investigated the production and characterization of membrane vesicles (MVs) in liquid cultures of S. coelicolor M145 from a structural and biochemical point of view using microscopic, physical and -omics analyzes. Two main populations of MVs, F3 and F4 MVs, with different size and cargo were isolated and purified. S. coelicolor MV cargo is complex and contains many “messages” such as proteins and metabolites. A total of 166 proteins involved in cell metabolism and differentiation, molecular processing and transport was identified in MVs. In particular, a subset of these proteins was protected from the degradation after proteinase K treatment, indicating their localization inside the vesicles. Vesicle proteome includes many stress response proteins which also play an important role in S. coelicolor morpho- physiological differentiation. Moreover, MVs packaged with an array of metabolites such as antibiotics, vitamins, amino acids and components of carbon metabolism. The analysis of S. coelicolor MV cargo will provide informations to elucidate their biogenesis and functions
