Murine polyploid trophoblast giant cells (TGCs) from day E9.5 implantation sites (experimental) vs E9.5 diploid embryos (control)

ABSTRACT: Array-based quantitative genome hybridization (aCGH) was performed to quantify gene copy number across the euchromatic portion of the genome of polyploid TGCs relative to embryonic diploid control cells. DNA was prepared from diploid embryonic cells and purified polyploid TGCs, labeled, and hybridized to microarrays. Experiment was performed in two biological replicates.

Throughout the plant and animal kingdoms specific cell types become polyploid, increasing their DNA content to attain a large cell size. In mammals, megakaryocytes (MKs) become polyploid before fragmenting into platelets. The mammalian trophoblast giant cells (TGCs) exploit their size to form a barrier between the maternal and embryonic tissues. The mechanism of polyploidization has been investigated extensively in Drosophila, in which a modified cell cycle--the endocycle, consisting solely of a  ...[more]
