Reproductive status alters transcriptomic response to infection in female Drosophila melanogaster

Project description:In this analysis, we used microarrays to contrast genome-wide transcript levels in virgin versus mated females before and after infection. We repeated the entire experiment using female mutants that do not form a germline. We found that multiple genes involved in egg production show reduced expression in response to infection, and that this reduction is stronger in virgins than it is in mated females. In germline-less females, expression of egg-production genes was predictably low and not differentially affected by infection. We also identified several immune responsive genes that are differentially induced after infection in virgins versus mated females.

A male-specific steroid hormone controls female reproductive biology in the malaria mosquito

Project description:Insects, unlike vertebrates, are generally believed to lack steroid hormones with functions predominantly associated with adult male biology. In the malaria mosquito Anopheles gambiae, the ecdysteroid 20-hydroxyecdysone (20E) appears to both control egg development in females and induce mating refractoriness and oviposition when sexually transferred by males. Here we show that these sex-specific functions are instead carried out by distinct steroids. We identify a male-specific oxidized form of 20E (3D20E) that upon sexual transfer switches off female mating receptivity, ensuring male paternity. Endogenous female 20E does not induce mating refractoriness, while it triggers oviposition in mated females when expression of a 20E-inhibiting kinase is repressed. 3D20E and 20E have different downstream targets, with 3D20E inducing expression of a tolerance factor that preserves female fitness during Plasmodium infection. The evolution of this male steroid has therefore not only shaped the mating biology of An. gambiae, but also impacted malaria transmission.

Developmental gene expression profiles of the human pathogen Schistosoma japonicum

Project description:The life cycle of schistosomes is complex, being characterised by a series of distinct parasitic and free-living phases involving an invertebrate snail host, water and a mammalian host. A custom designed oligonucleotide microarray was utilized to profile developmental gene expression in the Asian blood fluke, Schistosoma japonicum during these parasitic and free-living transitions. Total RNAs were isolated from lung schistosomula, juvenile females and males (paired but little or no egg production), adult males and females (paired with full scale egg production), eggs, miracidia, sporocysts and cercariae. We focused on the three distinct environmental phases of the lifecycle - aquatic/snail (eggs, miracidia, sporocysts, cercariae), juvenile (lung schistosomula and paired pre-egg laying adults) and adult (paired males and females, both examined separately) stages. Single colour schistosome life cycle with technical replicates in triplicate: Separate adult female and male worms at 4, 6, 6.5 and 7 weeks; cercariae; eggs; miracidia; schistosomula; and sporocysts. This study includes 6 adult male biological replicates.

Sex-specific gene expression profiles of Aedes aegypti 24 hr pupal heads

Project description:Custom microarrays were used to examine global differences in female vs. male gene expression in the developing pupal head of the dengue vector mosquito Aedes aegypti. RNA was extracted from the heads of male and female 24 hr pupae. 20 male or female heads were pooled for each of four replicates. Hybridization experiments were performed on the Nimblegen Aedes aegypti 12-plex microarray design: 090305_Aedes_aegypti_TEfam_expr.ndf. Four unique replicates and two repeat replicates were assessed in the hybridization experiment.

Deciphering life history transcriptomics in different environments

Project description:Investigation of whole genome transcription expression level changes in Drosophila mojavensis wild-type populations (Las Bocas:LB and Punta Prieta:PP). The experiment was designed to investigate life history transcriptomics in different environments. A total of 172 hybridizations were performed in this entire experiment. We used 135K 12-plex NimbleGen arrays. Total RNA was recovered from each sample listed below. The experimental design consisted a total of two populations (Las Bocas:LB; Punta Prieta:PP), two host diets (Agria:AG and Organ pipe:OP) and fourteen life stages (6 hr egg:E6; 1st instar larvae:L1; 2nd instar larvae:L2; 3rd instar larvae:L3; Early pupal stage:EP; Late pupal stage:LP; 0 Day old adult:0D; 3 Day old adult:3D; 4 Day old adult: 4D; 6 Day old adult:6D; 10 Day old adult:10D; 14 Day old adult:14D; 18 Day old adult:18D & 24 Day old adult:24D). Each chip measures the expression level of 14528 transcripts. Two to 5 replicates were used for each type (R1, R2, R3 etc.).
