Gene Expression Profile of Deletion RAD50, MRE11, XRS2

ABSTRACT: Deletion mutants of the yeast MRX complex (Rad50, Mre11 and Xrs2) were studied for gene expression levels. Significant difference in expression levels was observed in genes pertaining to DNA damage, cell cycle etc. as compared to the BY4741 wild type. Expression analysis with Phospholipase C over expression

Project description:The regulation of phospholipid biosynthesis in Saccharomyces cerevisiae through cis-acting upstream activating sequence inositol (UASino) and trans-acting elements, such as the INO2-INO4 complex and OPI1 by inositol supplementation in growth is thoroughly studied. In this study, we provide evidence for the regulation of lipid biosynthesis by phosphatidylinositol-specific phospholipase C (PLC) through UASino and two trans-acting elements. Gene expression analysis and radiolabelling experiments demonstrated that the overexpression of rice PLC in yeast cells altered phospholipid biosynthesis at the levels of transcriptional and enzyme activity. There are two biological replicates two hour and five hour induction with galactose. The genome wide expression of Saccharomyces cerevisiae strain BY4741 harbouring rPLC-pYES2 and a vectro control (pYES2) were compare with each other. A role for Snf2 related nucleosome spacing enzymes in genome-wide nucleosome organization

Project description:The positioning of nucleosomes within the coding regions of eukaryotic genes is aligned with respect to transcriptional start sites. This organization is likely to influence many genetic processes, requiring access to the underlying DNA. Here we show that the combined action of Isw1 and Chd1 nucleosome spacing enzymes is required to maintain this organization. In the absence of these enzymes regular positioning of the majority of nucleosomes is lost. Exceptions include the region upstream of the promoter, the +1 nucleosome and a subset of locations distributed throughout coding regions where other factors are likely to be involved. These observations indicated that ATP-dependent remodeling enzymes are responsible for directing the positioning of the majority of nucleosomes within the Saccharomyces cerevisiae genome. Gene expression in response to exposure to nitric oxide

Project description:To analyze gene expression pattern in response to nitric oxde in Saccharomyces cerevisiae, we performed microarray analysis using yeast cells treated with a nitric oxide donor S-nitroso-N-acetyl-DL-penicillamine (SNAP). S. cerevisiae S288c strain was cultured in synthetic dextrose (SD) minimal medium up to log phase (OD600 = 1), and then treated with 100 micro molar of SNAP for 10, 30, or 60 min. After treatment with SNAP, cells were disrupted adn total RNA was extracted. We commissioned following experiments to BIO MATRIX RESEARCH Inc. Nitric oxide-induced gene expression in S. cerevisiae was measured at 10, 30, or 60 minutes after exposure to 100 micro molar of nitric oxide donor SNAP.
