Gene expression profile in response to SDH2 deletion during filament formation in both Spider medium and Spider medium supplemented with 100mM glucose

ABSTRACT: Transcriptional profiling of Candida albicans comparing SDH2 deletion mutant cells with the wild-type cells in both Spider medium and Spider medium supplemented with 100mM glucose The SDH2 deletion mutant sdh2Î/Î and the wild-type strain SC5314 were used to perform the microarray experiments. Two-condition experiments: sdh2Î/Î vs SC5314 in Spider midium and sdh2Î/Î vs SC5314 in Spider midium supplemented with 100mM glucose. Biological replicates: 3 SDH2 deletion mutant sdh2Î/Î samples (test group), 3 wild-type strain SC5314 samples (control group), independently grown and harvested. One replicate per array.

Developmental stage-specific gene expression in the two-spotted spider mite (Tetranychus urticae)

Project description:We generated 77-bp Illumina reads from single messenger RNA libraries from four diverse developmental stages of the two-spotted spider mite to maximally capture the complement of transcribed sequences across development. Adult, nymphal, larvae and embryonic stages were separated using sieves of various pore sizes, and mites of various developmental stages were carefully selected for transcriptome library preparation. Samples were a mix of males and females to capture male and female patterns of transcription, and were reared on beans (Phaseolus vulgaris cv California Red Kidney). The RNA-Seq data was used for validation of gene models predicted by EuGene, and to study patterns of gene expression across development. Gene expression for spider mites from adult, nymph, larvae and embryonic developmental stages was examined (technical replicates were generated).

Genes Differentially Expressed as a result of Candida albicans SC5314 biofilms exposure to 10b

Project description:Biomaterial infections are an increasingly alarming problem, and because of their intrinsic recalcitrance to conventional therapy, a new class of antifungal drugs must be explored. 10b, a 2-aminotetralin derivate, was synthesized as a novel chemical structural antifungal agent and exibited strong anti-biofilm activity. To further investigate the action mechanism, we used microarray analysis to investigate the genes expression profiles of C. albicans biofilms treated or untreated with 10b and found 150 genes were differentially expressed. Of them, 69 showed a decrease in expression and 81 showed an increase in expression -10 differentially expressed genes related to biofilm formation, Filamentous or hypha growth. A gene related to specifically hydrolyzing Î²-1, 3 glucan was significantly increased. 10 down-regulated genes were involved in glycolysis, fermentation and active oxygen scavenging. 15 overexpressed genes were related to the lipid metabolic process. Of them, 13 genes were directly linked to ergosterol biosynthesis including ERG2, ERG6 and ERG11. 10 genes related to translation were over-expressed. Among them, 2 genes involved in negative regulation of transcription were significantly up-regulated. Total RNA from the control SC5314 biofilms and 10b-treated SC5314 biofilms were used to generate target cDNA, and then hybridized to 8k Candida albicans Genome Array Genechips, representing about 7925 characterized Candida albicans genes. Two independent experiments were conducted. Reference strain was control SC5314 biofilms and test strain was SC5314 biofilms treated with 10b.

Examination of small RNA from spider mites of adult, embryonic and pooled larval/nymphal developmental stages.

Project description:Our goal was to investigate the transcriptional response of T. urticae to different Arabidopsis plants with varying levels of indole glucosinolates. Bean is the ancestral host plant for the T. urticae strain used in this study. Adult female spider mites were transferred to the three Arabidopsis lines (Col-0, qKo, atr1D) or bean, and after 24 hours, the gene expression levels were analyzed. 12 samples were investigated; 4 biological replicates for three comparisons: QKO (qKo vs. Col-0), ATR1D (atr1D vs. Col-0), and COL-0 (Col-0 vs. bean).

Host plant shift and acaricide resistance effects on gene expression in the two-spotted spider mite, T. urticae

Project description:The goal of our microarray experiments was twofold: 1) Compare the gene expression profile of acaricide resistant spider mite strains (MAR-AB and MR-VP) with that of a susceptible spider mite strain (London); 2) Study gene expression changes in spider mites from the London strain upon transfer from bean, a suitable host, to tomato, a less favorable host. These gene expression changes upon host change were measured for three timepoints (2 hour on tomato (Tomato-2h), 12 hour on tomato (Tomato-12h) and 5 generations on tomato (Tomato-5G)). 23 samples were analyzed: 6 biological replicates for MR-VP, 5 biological replicates for MAR-AB and four biological replicates each for Tomato-2h, Tomato-12h and Tomato-5G

Tetranychus urticae secretome - The Salivary Protein Repertoire of the Polyphagous Spider Mite Tetranychus urticae: A Quest for Effectors

Project description:The two-spotted spider mite Tetranychus urticae is an extreme polyphaguous crop pest. Next to an increased detoxification potential of plant secondary metabolites, it has recently been shown that spider mites manipulate plant defences. Salivary constituents are proposed to play an important role during the interaction with its many hosts. The proteomic composition of saliva delivered into artificial diet by spider mites adapted to various hosts - bean, soy, maize, tomato -was determined using Orbitrap mass spectrometry. Over 200 different proteins were identified, many of unknown function and in numerous cases belonging to multi-membered gene families. A selection of these putative salivary proteins was validated using whole-mount in situ hybridizations and expression was shown to be localized in the anterior and dorsal podocephalic glands of the spider mite. Host-plant dependent expression was evident from the proteomics dataset and was further studied in detail by micro-array based genome wide gene expression profiling of mites maintained on the host plants under study. Previously obtained gene-expression datasets were further used to get more insight in the expression profile over different life stages and physiological states. To conclude, for the first time the T. urticae salivary proteome repertoire was characterized using a custom feeding hemisphere-based enrichment technique. This knowledge will assist in unraveling the molecular interactions between phytophagous mites and their host plants. This may ultimately facilitate the development of mite-resistant crops.

Biofilm strains (48 hour biofilms) were compared to planktonic strains (log phase planktonic cells) in Spider medium at 37C.
