Transcriptional Response of Mycobacterium tuberculosis CDC1551 to treatment with 140 mM NaCl for 1 hr

ABSTRACT: To identify mediators of the osmotic stress response in Mycobacterium tuberculosis (Mtb), we performed transcriptional profiling of WT CDC1551 following treatment with 140 mM NaCl for 1 hr relative to an untreated control. 140 mM NaCl was chosen to reflect the approximate osmolarity of human plasma (i.e., 280 mOsm/L), which may be relevant during the course of infection. CDC1551 was treated with 140 mM NaCl for 1 h and compared to the same strain treated with 0 mM NaCl for 0 h. 4 biological replicates, independently grown and harvested. One replicate per array.

Project description:rs06-07_della - della-regulation of salt stress responses - Identification of DELLA-dependent dowtream targets in response to salt stress - Aim was to determine downstream target of DELLA proteins involved in salt stress tolerance. Wt, ga1-3, penta seeds were sterilized, sown on MS agar plates then put for stratification for 3 days at 4degreeC. Plates were placed in growth cabinet for 9 days. Seedlings were transferred to 24 well-plates, with 2 seedlings per well (0.5 ml MS liquid per well). Plates were placed in the same condition for 3 days. Finally, NaCl were added (final concentration 200 mM), except for the control. The salt treatment was applied for 30 min and 1h. Treatment was stopped by freezing in liquid nitrogen. Protein interaction AP-MS data for TAP-CSAG1 and TAP-CSAG2 from HEK293 cells

Project description:Ten 15 cm2 plates of HEK293 cells stably expressing TAP-vector, TAP-CSAG1, or TAP-CSAG2 were lysed with TAP lysis buffer (10% (v/v) glycerol, 50 mM HEPES-KOH pH 7.5, 100 mM KCl, 2 mM EDTA, 0.1% (v/v) NP-40, 10 mM NaF, 0.25 mM Na3VO4, 50 mM β-glyerolphosphate, 2 mM DTT, and 1X protease inhibitor cocktail (Sigma)) and cleared supernatants were bound to IgG-Sepharose beads (GE Amersham) and then washed in lysis buffer and TEV buffer (10 mM HEPES-KOH pH 8.0, 150 mM NaCl, 0.1% (v/v) NP-40, 0.5 mM EDTA, 1 mM DTT, and 1X protease inhibitor cocktail). Protein complexes were cleaved off the beads by TEV protease (Sigma) and incubated with calmodulin-Sepharose beads (GE Amersham) in calmodulin binding buffer (10 mM HEPES-KOH pH 8.0, 150 mM NaCl, 1 mM Mg acetate, 1 mM imidazole, 0.1% (v/v) NP-40, 6 mM CaCl2, 10 mM 2-mercaptoethanol) and then washed in calmodulin rinse buffer (50 mM Ammonium bicarbonate pH 8.0, 75 mM NaCl, 1 mM Mg Acetate, 1 mM Imidazole, 2 mM CaCl2) before eluted with SDS sample buffer, subjected to SDS-PAGE, and stained with GelCode Blue stain (Thermo Fisher Scientific) before protein identification by LC-MS/MS.
