Transcriptional Response of Mycobacterium tuberculosis CDC1551 Rv0516c::Tn to treatment with 140 mM NaCl for 1 hr

ABSTRACT: To characterize the role of Rv0516c in the osmotic stress response of Mycobacterium tuberculosis (Mtb), we performed transcriptional profiling of CDC1551 Rv0516c::Tn following treatment with 140 mM NaCl for 1 hr relative to an untreated control. CDC1551 Rv0516c::Tn was treated with 140 mM NaCl for 1 h and compared to the same strain treated with 0 mM NaCl for 0 h. 4 biological replicates, independently grown and harvested. One replicate per array.

Project description:To identify mediators of the osmotic stress response in Mycobacterium tuberculosis (Mtb), we performed transcriptional profiling of WT CDC1551 following treatment with 140 mM NaCl for 1 hr relative to an untreated control. 140 mM NaCl was chosen to reflect the approximate osmolarity of human plasma (i.e., 280 mOsm/L), which may be relevant during the course of infection. CDC1551 was treated with 140 mM NaCl for 1 h and compared to the same strain treated with 0 mM NaCl for 0 h. 4 biological replicates, independently grown and harvested. One replicate per array.

Transcritpome analysis of Mycobacterium tuberculosis (Mtb) mutants deficient in ergothioneine, or mycothiol production

Project description:This experiment was performed to determine deferentially regulated genes in the absence of redox buffer ergothioneine (EGT), or mycothiol (MSH). We had used two Mtb mutants namely egtA:Tn and egtD:Tn which are deficient in EGT production ( Transposon Mutant of EGT biosynthesis pathway); In addition, we also used Mtb mshA mutant, which does not produce MSH ( Deletion mutant of Mycothiol biosynthesis ). For microarray analysis, RNA was obtained from exponentially growing (OD 600 nm - 0.6-0.8) CDC1551 (wild type), mshA mutant, egtA:Tn and egtD:Tn strains cultured in 7H9 medium supplemented with ADST (Albumin Dextrose Saline Tyloxapol) using RNApro (MP Biomedicals, USA) as per manufacturerâs instructions. The quality of RNA was examined by running the samples on Experion gene chips (BioRad).

Project description:Ten 15 cm2 plates of HEK293 cells stably expressing TAP-vector, TAP-CSAG1, or TAP-CSAG2 were lysed with TAP lysis buffer (10% (v/v) glycerol, 50 mM HEPES-KOH pH 7.5, 100 mM KCl, 2 mM EDTA, 0.1% (v/v) NP-40, 10 mM NaF, 0.25 mM Na3VO4, 50 mM β-glyerolphosphate, 2 mM DTT, and 1X protease inhibitor cocktail (Sigma)) and cleared supernatants were bound to IgG-Sepharose beads (GE Amersham) and then washed in lysis buffer and TEV buffer (10 mM HEPES-KOH pH 8.0, 150 mM NaCl, 0.1% (v/v) NP-40, 0.5 mM EDTA, 1 mM DTT, and 1X protease inhibitor cocktail). Protein complexes were cleaved off the beads by TEV protease (Sigma) and incubated with calmodulin-Sepharose beads (GE Amersham) in calmodulin binding buffer (10 mM HEPES-KOH pH 8.0, 150 mM NaCl, 1 mM Mg acetate, 1 mM imidazole, 0.1% (v/v) NP-40, 6 mM CaCl2, 10 mM 2-mercaptoethanol) and then washed in calmodulin rinse buffer (50 mM Ammonium bicarbonate pH 8.0, 75 mM NaCl, 1 mM Mg Acetate, 1 mM Imidazole, 2 mM CaCl2) before eluted with SDS sample buffer, subjected to SDS-PAGE, and stained with GelCode Blue stain (Thermo Fisher Scientific) before protein identification by LC-MS/MS.
