Project description:Bovine tuberculosis (bTB), caused by Mycobacterium bovis (M. bovis), poses a signi!cant global health and economic burden. Despite extensive research, a comprehensive understanding of M. bovis pathogenesis, particularly its transcriptional adaptation across different growth phases and within the host environment, remains incomplete. Here, we performed a comprehensive transcriptomic analysis of virulent M. bovis and the attenuated M. bovis BCG strain (BCG) across early-log, mid-log, and stationary growth phases to elucidate the molecular underpinnings of their phenotypic distinctions. RNA sequencing revealed distinct gene expression pro!les, highlighting signi!cant differences in pathways related to cell wall biosynthesis, lipid metabolism, transcriptional regulation, protein secretion, and the PE/PPE protein family. Notably, the virulent M. bovis exhibited an upregulation of virulence-associated genes, especially those involved in lipid metabolism and secretion systems, during the transition from early- and mid-log phases to the stationary phase. In contrast, BCG demonstrated an enrichment in stress response and metabolic adaptation pathways during the similar transition. Gene network analysis of differentially expressed genes (DEGs) provided a systems-level view of the transcriptional programs governing M. bovis and M. bovis BCG physiology, identifying !ve key modules of co-expressed genes that regulate small molecules transport, amino acid biosynthesis and immune evasion in M. bovis. Furthermore, we analyzed M. bovis transcriptional responses during murine lung infection, identifying a core set of DEGs linked to host-pathogen interactions and mechanisms of persistence. These !ndings offer novel insights into M. bovis adaptation strategies during survival under de!ned, in vitro growth conditions and highlight potential targets for improved vaccine design and therapeutic interventions following further analyses during host infection.

Following infection, the pathogen resides and persists inside host macrophages by subverting host immune responses via a diverse range of mechanisms. Here, a high-density bovine microarray platform was used to examine the bovine monocyte-derived macrophage transcriptome response to M. bovis infection relative to infection with the attenuated vaccine strain, M. bovis Bacille Calmette–Guérin. Differentially expressed genes were identified (adjusted P-value ≤ 0.01) and interaction networks generated across an infection time course of 2, 6 and 24 h. The largest number of biological interactions was observed in the 24 h network, which exhibited small-worldscale-free network properties. The 24 h network featured a small number of key hub and bottleneck gene nodes, including IKBKE, MYC, NFKB1 and EGR1 that differentiated the macrophage response to virulent and attenuated M. bovis strains, possibly via the modulation of host cell death mechanisms. These hub and bottleneck genes represent possible targets for immunomodulation of host macrophages by virulent mycobacterial species that enable their survival within a hostile environment.

Project description:Mycoplasma bovis (M.bovis) is a critical pathogen of bovines resulting in pneumonia, mastitis, arthritis, etc. To reveal its virulence related factors, a virulent M. bovis HB0801 and its derived vaccine strain P150 were infected calve and the transcriptome profiles of PBMCs were compared by using of the microarray at 7 days after infection. The data postulated the pathogenic mechanism of wild strain and immune mechanism of the attenuated strain to provide clues for the further research of the interaction between M. bovis and its host.

The aims were two-fold, namely to define biosignatures that could predict vaccine success before challenge, and biomarker patterns that correlated with anamnestic protective responses following exposure to virulent M. bovis. Further, Our aim was to define these markers to be detectable without in vitro antigenic challenge. After BCG vaccination, we defined a specific pulmonary gene expression signature related to the connective tissue development and function network that predicted vaccine success before M. bovis challenge. In addition, a Th17-related cytokine profile was found that correlated with vaccine-induced protective immunity following infection with virulent M. bovis in the lung as well as additional genes that were up-regulated in the spleens of vaccinated animals post-infection that was related to neutrophil biology and inflammation. This study has therefore prioritized both biomarkers predicting vaccination success before challenge and bio-signatures that are associated with protective immune responses that will be useful to evaluate future vaccine candidates. Two groups of 20 mice each were immunised by a single intradermal injection of 2 x 10 to 5 CFU of M. bovis BCG (Vaccinated), or Hanks buffered salt solution (HBSS) (Unvaccinated). Six weeks later 5 mice from each group were euthanized for immunological analyses and the remaining mice from each group were challenged with approx 600 CFU M. bovis via the intranasal route. At days 3 and 14 post challenge five mice per group were euthanized and spleens and lungs harvested.

Project description:Bovine babesiosis is a tick-borne disease that poses a significant economic threat to cattle industries in tropical and subtropical areas, and Babesia bovis is the most virulent causative agent of bovine babesiosis. This apicomplexan parasite infects erythrocytes of cattle causing severe hemolytic disease, and animals that survive acute infections become persistently infected for life. Young cattle (< 6 months of age) are resistant to infection while adult cattle (>1 year of age) are highly susceptible and succumb to acute infection; however, the immunological mechanisms associated with the age-resistance remain unclear. Protective host immunity involves peripheral blood mononuclear cells (PBMCs) including natural killer (NK) and T cells, and activated macrophages that act to control the pathogen. In this study, we examined the transcriptional signatures of PBMCs from adult cattle (>1.5 years old) experimentally infected with the B. bovis virulent strain S74-T3Bo. Transcriptional signatures evident during the acute phase of babesiosis were categorized into immune-related and non-immune genes. We identified both upregulated and downregulated genes, with fold changes ranging from 2 to 263-fold. We discuss our findings in the context of immune responses to acute disease as a mechanism for adult host survival, with a focus on the molecular functions and biological processes involved in the response to B. bovis infection. In this RNA-Seq analysis, we identified genes related to the immune system, including cytokines and chemokines, complement, cell signaling pathways and surface molecules that may play a role in the recognition of pathogen-associated molecular patterns. In addition, non-immune genes potentially involved in cell proliferation, cell migration, development, energy production, protein-protein interactions, molecular transport, and flagella assembly were also identified.

Project description:Infection of cattle with Mycobacterium bovis causes severe financial hardship in many countries, in addition to presenting a health risk for humans. As an intracellular pathogen, M. bovis, adapted to survive and thrive within the intramacrophage environment. However, little is known about expression patterns in the macrophage, particularly in the bovine host. In this study, DNA microarray analysis was used to detect genes expressed in Holstein bovine macrophages derived from peripheral blood mononuclear cells infected during four hours with two Argentinean strains of M. bovis, a virulent strain, 04-303 and an attenuated strain, 534. Genes encoding antrax toxin receptor, cell division and apoptosis regulator, ankyrin proteins that are found within the membrane associated cytoskeleton, protein of cell differentiation and regulators of endocytic traffic of membrane were more strongly expressed in infected macrophages.
