Effect of MCMV infection on gene expression in ovaries of pregnant and non-pregnant mice

ABSTRACT: Affymetrix Gene ST 1.0 arrays were performed on total RNA obtained from ovaries of 5 female mice (pregnant or non-pregnant; mock or MCMV infection). Mice were infected 9.5 days post conception with 2x10e05 PFU MCMV i.v.. RNA was prepared at 5 days post infection (14.5 days post conception). Total RNA prepared from ovaries of 5 mice per condition was pooled and subjected to microarray analysis. Data were normalised using RMA.

The ISWI ATPase Snf2L is required for superovulation and regulates Fgl2 in differentiating mouse granulosa cells

Project description:We investigate the role of Snf2l in ovaries by characterizing a mouse bearing an inactivating deletion on the ATPase domain of Snf2l (Ex6DEL). Snf2l mutant mice produce significantly fewer eggs than control mice when superovulated. Thus, gonadotropin stimulation leads to a significant deficit in secondary follicles and an increase in abnormal antral follicles. We profiled the expression of granulosa cells from Snf2l WT and Ex6DEL mice treated with pregnant mares' serum gonadotropin followed by human chorionic gonadotropin Granulosa cells from either Snf2l WT or Ex6DEL mice treated with PMSG followed by hCG were collected at 0h and 4h post-hCG. Each array includes granulosa cells pooled from 5 mice.

Transcription profiling of rat heart, kidney and placenta from fetuses obtained from dams with diet-induced intra-uterine growth restriction vs. normal

Project description:Pregnant rats were randomly divided into two groups. Dams were then housed individually and fed throughout gestation a 22 % protein diet, representing the control group (C, n=5), or an 9% isocaloric protein diet, representing the low-protein (LP, n=6). Both diets were isocaloric, because the protein deficiency in the LP diet was compensated by the addition of carbohydrates. Feeding of LP diet began on the day of conception and continuing throughout pregnancy. Transcription profiling of pig endometrium from pregnant and non-pregnant sows

Project description:In an attempt to unveil part of the molecular processes controlling porcine placentation we have investigated the pregnancy induced gene expression in the porcine endometrium at Day 14 after insemination using the Affymetrix GeneChip® Porcine Genome Array. Experiment Overall Design: At Day 14 after insemination, samples were obtained from the endometrium of pregnant sows and sows inseminated with inactivated semen for RNA extraction and hybridization on Affymetrix microarrays. The uteri were removed and each uterine horn was flushed with PBS containing 1% fetal calf serum, and subsequently opened longitudinally at the anti-mesometrial side. The sites of embryonic attachment were macroscopically visual in the endometrium on the mesometrial side in form of hyperemic zones. Samples of the endometrium (lamina epithelialis, lamina propria and tela submucosa, but not tunica muscularis) were isolated from proximal (the end close to the ovaries), intermedial, and distal (next to the corpus uteri)) sections of each uterine horn. Samples were taken from the endometrium of the non-pregnant animals at comparable locations.

Transcriptome profile form endometrial biopsy samples of pregnant and non-pregnant cows

Project description:Purpose: characterize the uterine transcriptome profiles of pregnant (P) versus non-pregnant (NP) cows during early pregnancy and attempted to define a potential set of marker genes that can be valuable for predicting pregnancy outcome. Methods: beef cows were synchronized and artificially inseminated at detected estrus. Six days after AI, jugular blood samples and a biopsy from the uterine horn contralateral to the ovary containing the corpus luteum were collected. Based on pregnancy outcome on day 30, samples were retrospectively allocated to the following groups: Pregnant and Non-Pregnant. Both groups had similar plasma progesterone concentrations on D6. Uterine biopsies were submitted to RNA-Seq analysis in a Illumina HiScanSQ platform. Results: The 272,685,768 million filtered reads were mapped to the Bos taurus reference genome and 14,654 genes were analyzed for differential expression between groups. Transcriptome data showed that 216 genes are differently expressed when comparing NP versus P uterine tissue (Padj≤0.1). More specifically, 36 genes were up-regulated in P cows and 180 are up-regulated in NP cows. Conclusions: this study characterized a unique set of genes, expressed in the uterus on 6 days after insemination, that indicate a receptive state leading to pregnancy success. Identifying estrogen regulated gene expression profile of pre-implantation mouse embryos

Project description:Comparison of the gene expression profiles of pre-implantation embryos at day 3.5 post coitum from normal pregnant mice (control); embryos from mice treated with ICI (specific estrogen receptor inhibitor); and embryos in the oviduct that were blocked from entering the uterus by ligation. Results provide insight into the function of estrogen regulated genes and uterine factors involved in the early implantation process. RNA were extracted from 3 groups of 100-120 embryos (a) embryos at d3.5 from uterus of normal pregnant mice; (b) embryos at d3.5 from uterus of ICI treated mice; and (c) embryos at d3.5 from the ligated oviduct.

RNA-seq of pancreatic islets from pregnant mice

Project description:During pregnancy, the energy requirements of the fetus impose changes in maternal metabolism. Increasing insulin resistance in the mother maintains nutrient flow to the growing fetus, while prolactin and placental lactogen counterbalance this resistance and prevent maternal hyperglycemia by driving expansion of the maternal population of insulin-producing beta-cells. However, the exact mechanisms by which the lactogenic hormones drive beta-cell expansion remain uncertain. Here we show that serotonin acts downstream of lactogen signaling to drive beta-cell proliferation. Serotonin synthetic enzyme Tph1 and serotonin production increased sharply in beta-cells during pregnancy or after treatment with lactogens in vitro. Inhibition of serotonin synthesis by dietary tryptophan restriction or Tph inhibition blocked beta-cell expansion and induced glucose intolerance in pregnant mice without affecting insulin sensitivity. Expression of the Gq-linked serotonin receptor Htr2b in maternal islets increased during pregnancy and normalized just prior to parturition, while expression of the Gi-linked receptor Htr1d increased at the end of pregnancy and postpartum. Blocking Htr2b signaling in pregnant mice also blocked beta-cell expansion and caused glucose intolerance. These studies reveal an integrated signaling pathway linking beta-cell mass to anticipated insulin need during pregnancy. Modulators of this pathway, including medications and diet, may affect the risk of gestational diabetes.
