SiRNA profiling of human H157 non-small cell lung carcinoma cells with and without DJ-1 expression knocked-down


ABSTRACT: DJ-1 expression protects H157 cells from cell death, using siRNA oligomers we knocked down DJ-1 and evaulated gene expression using Affymetrix U133A GeneChip arrays. Experiment Overall Design: H157 cells were transfected every 24 hours with siRNA oligomers for 3 consecutive days, lysed 96 hours after the first transfection, and total RNA extracted. cRNA was generated and used to probe Affymetrix U133A GeneChip microarrays.

DJ-1/PARK7, a cancer- and Parkinson's disease (PD)-associated protein, protects cells from toxic stresses. However, the functional basis of this protection has remained elusive. We found that loss of DJ-1 leads to deficits in NQO1 [NAD(P)H quinone oxidoreductase 1], a detoxification enzyme. This deficit is attributed to a loss of Nrf2 (nuclear factor erythroid 2-related factor), a master regulator of antioxidant transcriptional responses.
