Project description:Fast and accurate identification of pathogenic bacteria along with the identification of antibiotic resistance proteins is of paramount importance for the treatment of patients and public health. While mass spectrometry has become an important technique for these purposes there is a lack of mass spectrometry workflow offering this capability. To meet this need we have augmented the previously published Microorganism Classification Identification (MiCId) workflow with this capability. Evaluation results showed that MiCId's workflow has a sensitivity value around 86% and a precision greater than 95% in the identification of antibiotic resistance proteins. Futhermore, we showed that MiCId's workflow is fast. It is capable of providing microorganismal identification, protein identification, sample biomass estimation, and antibiotic resistance protein identification in about 6-17 minutes using computer resources that are available in most desktop and laptop computers making it highly portable workflow. Project description:We wanted to measure the transcriptomic changes of P.aeruginosa (PAO1 strain) when treated with plant extracts (Melastoma dodecandrum Lour, and Potentilla indica) and antibiotics (triclosan, rifampicin, gentamicin and ciprofloxacin) against DMSO control to aid in understanding the mode of action of our plant extracts. PAO1 samples were treated with plant extracts and antibiotics at MIC, for 24h at 37°C with OD readings taken at 600 nm every 10 mins using a microplate reader.
Project description:Staphylococcus aureus is one of the most important pathogens in humans and animals, multiply resistant strains are increasingly widespread, new agents are needed for the treatment of S. aureus. Rhein, a natural plant product, has potential antimicrobial activity against Staphylococcus aureus. We employed Affymetrix Staphylococcus aureus GeneChipsTM arrays to investigate the global transcriptional profiling of Staphylococcus aureus ATCC25923 treated with rhein. Results provided insight into mechanisms involved in rhein - Staphylococcus aureus interactions.
