We performed total RNA sequencing using whole blood cells collected from multiparous Holstein Friesian dairy cows with naturally occurring mastitis to investigate the changes in systemic gene expression and their association with inflammatory responses. Project description:The immune response associated with mastitis caused by Mycoplasma bovis is a very complicated biological process in several type of cells, including immune cells, mammary epithelial cells and, endothelial cells. Thus, revealing of the microRNAs in the Mycoplasma bovis infected mammary gland tissues is particularly important for the immune response mechanism to Mycoplasma bovis. Firstly, mammary gland tissue samples were collected from Holstein cows and screened for Mycoplasma bovis. Then, total RNA was isolated from mycoplasma bovis infected tissues and RNA sequencing was performed. After bioinformatics analysis, GO and KEGG analysis of target genes of identified microRNAs were conducted. Our results revaled that 24 of the known microRNAs were expressed differently and 13 of the novel microRNAs were expressed differently in Mycoplasma bovis positive tissues. The target genes of these microRNAs were found to be associated with especially inflammation pathways. In conclusion, this study demonstrated that identified miRNAs may be involved in the signaling pathways during mastitis case caused by Mycoplasma bovis.

Project description:Milk protein is one of the most important economic traits in the dairy industry. Yet, the miRNA gene regulatory network for the synthesis of milk protein in mammary is poorly understood. In this study, the hypothesis was that miRNAs have potential roles in bovine milk protein production. Using miRNA-seq and RNA-seq, we investigated the miRNAs profiles of mammary glands from 12 Chinese Holstein cows with six cows at peak of lactation and six in non-lactating period, from which three cows were in high and three in low milk protein percentage.

The mouse differentially expressed genes through the use of comparative mapping/genomics and positional information on reported QTL for bovine mastitis allowed identifying 293 potential candidate genes for bovine mastitis. This study will enable other researchers to combine our mRNA expression data with genetic association studies to discover genomic variation underlying variation of susceptibility to mastitis in dairy cows. We measured levels of gene expression in healthy (saline injected) and diseased (E. coli injected) mammary gland tissue (treatment = 2, in sub-plots). The mammary tissue samples were collected at two-time points: 24 hr and 48 hr post-injection (time = 2, in main-plots). Each inbred mouse was injected with both treatments: the 4th left mammary gland was injected with saline solution and the 4th right mammary gland was injected with E. coli solution (mice = 4). The expression profiling was carried out using two biological replications (replication = 2) at each time point.

Project description:Purpose: To identify the expression profiling of miRNA in peripheral blood of dairy cows in response to S. aureus-infected mastitis and explore the biomarkers for early diagnosis of S. aureus-infected mastitis. Methods: RNAseq technology was used to determine the expression profiles of microRNA (miRNA) from peripheral blood of Chinese Holstein cows infected with S. aureus at 0, 1, 3, 5, and 7 days. Results: Ttal of 288 differentially expressed miRNAs (DIE-miRNA) including 108 known and 180 novel predicted miRNAs, involved in 10 immune system-related signaling pathways. Compare with the 0 dpi, the number of DIE-miRNAs in 1, 3, 5, and 7 dpi groups were 12, 21, 75, and 48, respectively. It was also found that the expression variation of up-regulated expression of miR-320a, miR-19a, and miR-19b as well as down-regulated expression of miR-143, miR‑205, and miR‑24 reached a significant level on the 5 dpi and 7 dpi. However, at different times after S. aureus infection, miR-1301 was significantly up-regulated in peripheral blood. miR-2284r was significantly down-regulated. Conclusion: miR-1301 and miR-2284r might be the new blood biomarkers for S. aureus-infected dairy cow mastitis. The above results laid a new foundation for the research and development of molecular diagnosis and biological therapy technology for S. aureus-infected mastitis in dairy cow.

Project description:Early detection of bovine subclinical mastitis may improve treatment strategies and reduce the use of antibiotics. Herein, individual milk samples from Holstein cows affected by subclinical mastitis induced by S. agalactiae and Prototheca spp. were analyzed by untargeted and targeted mass spectrometry approaches to assess changes in their peptidome profiles and identify new potential biomarkers of the pathological condition. Results showed a higher amount of peptides in milk positive at the bacteriological examination when compared with the negative control. However, the different pathogens seemed not to trigger specific effects on milk peptidome. The peptides that best distinguish positive from negative samples are mainly derived from the most abundant milk proteins, especially from β- and αs1-casein, but also include the antimicrobial peptide casecidin 17. These results provide new insights into the physiopathology of mastitis. Upon further validation, the panel of potential discriminant peptides could help to the development of new diagnostic and therapeutic tools.

Project description:Mastitis is one of the most prevalent and economically important diseases of dairy animals. The disease is caused by ascending bacterial infection through the teat canal. Among the most common mastitis-causing bacteria are gram-negative coliforms, gram-positive streptococci and staphylococci, and mycoplasma. The most prominent cellular hallmark of acute mammary infection is a massive recruitment of blood neutrophils into the tubular and alveolar milk spaces. The complex biological processes of leukocyte recruitment, activation, adhesion, and migration in the mammary gland remain largely elusive to date. While field research of mastitis in dairy animals contributed a lot to the development of mitigation, control, and even eradication programs, little progress was made toward understanding the molecular mechanisms underlying the pathogenesis of the disease. We report here experimental mastitis model systems in lactating mice challenged with field strains of common udder pathogens in dairy cows. We used these model systems to apply recently developed multiplex gene expression technology (Nanostring nCounter), which enabled us to study the expression of over 700 immune genes. Our analysis revealed a core of 100 genes that are similarly regulated and functionally or physically interacting in E. coli, M. bovis and Strep uberis murine mastitis. Common significantly enriched gene sets include TNFɑ signaling via NFkB, Interferon gamma and alpha response, and IL6-JAK-STAT3 signaling. In addition, we show a significantly enriched expression of genes associated with neutrophil extracellular traps (NET) in glands challenged by the three pathogens. Ligand-receptor analysis revealed interactions shared by the three pathogens, including the interaction of the cytokines IL1β, IL1ɑ, and TNFɑ with their receptors, and proteins involved in immune cell recruitment such as complement C3 and ICAM1 (with CD11b), chemokines CCL3 and CCL4 (with CCR1), and CSF3 (with CSF3R). Taken together, our results show that mammary infection with E. coli, M. bovis and Strep uberis culminated in the activation of a conserved core of immune genes and pathways including NET formation.
