MRNA analysis of gene expression in adipose tissue to compare high fat fed sgp130Fc transgenic vs IL6 knock-out vs wild-type mice

ABSTRACT: We want to analyse on a broad level the difference of gene expression in the adipose tissue between high fat fed sgp130Fc Transgenic which have no IL6 transsignalling and the IL6 knockout which have no IL6 signalling at all. Because the sgp130Tg mice show a marked adipogenic phenotype we want to compare it with the IL6 KO. Total RNA extracted from flash frozen adipose tissue from obese, 5hr-fasted mice.

Bio-ball vs mesoball

Project description:MIXL1-GFP reporter lines (HESC3MIXL1-GFP/w and MEL1MIXL1-GFP/w, referred to as HES3 and MEL1 in this submission) were differentiated as spin embryonic bodies (Ebs) supplemented with mesodermal inducing growth factors (BMP4,SCF, VEGF based). Methylcellulose hematopoietic colony forming assays were performed by culturing dissociated day 4 EBs in the presence of hematopoietic growth factors (VEGF, SCF, IL3, IL6, TPO, EPO and FLT3L) for 7-9 days. Addition of WNT3a to the methylcellulose lead to the formation of compact, mesodermal colonies, which we term 'mesosphere' (Meso-balls). We demonstrated that the inclusion of WNT3a in the methylcellulose could be replaced with BIO, a GSK3 inhibitor, acting as a canonical WNT signaling agonist. Mesospheres formed in the culture supplemented with BIO molecules were termed 'BIO-balls'. Expression profiling between 'meso-balls' and 'BIO-balls' were compared and analyzed for markers assisiting in defining their phenotypes.

Slow growing behavior in African trypanosomes during adipose tissue colonization

Project description:When Trypanosoma brucei parasites, the causative agent of sleeping sickness, colonize the adipose tissue, they rewire gene expression. Whether this adaptation affects population behavior and disease treatment remained unknown. By using a mathematical model, we estimate that the population of adipose tissue forms (ATFs) proliferates slower than blood parasites. Analysis of the ATFs proteome, measurement of protein synthesis and proliferation rates confirm that the ATFs divide on average every 12hr, instead of 6hr in the blood. Importantly, the population of ATFs is heterogeneous with parasites doubling times ranging between 5hr and 35hr. Slow-proliferating parasites remain capable of reverting to the fast proliferation profile in blood conditions. Intravital imaging shows that ATFs are refractory to drug treatment. We propose that in adipose tissue, a subpopulation of T. brucei parasites acquire a slow growing behavior, which contributes to disease chronicity and treatment failure.

DNA methylation analysis of subcutaneous adipose tissue abdominal versus subcutenaous adipose tissue gluteal

Project description:In humans, adipose tissue is distributed in subcutaneous abdominal and subcutaneous gluteal depots that comprise a variety of functional differences. Whereas energy storage in gluteal adipose tissue has been shown to mediate a protective effect, an increase of abdominal adipose tissue is associated with metabolic disorders. However, the molecular basis of depot-specific characteristics is not completely understood yet. Using array-based analyses of transcription profiles, we identified a specific set of genes that was differentially expressed between subcutaneous abdominal and gluteal adipose tissue. To investigate the role of epigenetic regulation in depot-specific gene expression, we additionally analyzed genome-wide DNA methylation patterns in abdominal and gluteal depots. By combining both data sets, we identified a highly significant set of depot-specifically expressed genes that appear to be epigenetically regulated. Interestingly, the majority of these genes form part of the homeobox gene family. Moreover, genes involved in fatty acid metabolism were also differentially expressed. Therefore we suppose that changes in gene expression profiles might account for depot-specific differences in lipid composition. Indeed, triglycerides and fatty acids of abdominal adipose tissue were more saturated compared to triglycerides and fatty acids in gluteal adipose tissue. Taken together, our results uncover clear differences between abdominal and gluteal adipose tissue on the gene expression and DNA methylation level as well as in fatty acid composition. Therefore, a detailed molecular characterization of adipose tissue depots will be essential to develop new treatment strategies for metabolic syndrome associated complications. DNA methylation profiles of abdominal adipose tissue (6 samples) and gluteal adipose tissue (6 samples) were generated using Infinium methylation 450K BeadChips from Illumina (Illumina, San Diego, USA).

Gene expression profiles of the human breast cancer cell line MDA-MB-231 co-cultured with adipose tissue from mice on chow or high-fat diet

Project description:Obesity is a known risk factor for breast cancer. To identify genes and underlying pathways in human triple-negative breast cancer cells affected by interaction with adipose tissue, MDA-MB-231 breast cancer cells were cultivated in a co-culture system with or without adipose tissue explants from mice for the purpose of a microarray gene expression analysis. Co-culture of MDA-MB-231 breast cancer cells was performed with adipose tissue explants obtained from C57BL/6J mice that were fed a high-fat diet (HFD, 58%Kcal from fat) or normal chow diet (NC; 11%Kcal from fat) ad libitum for 16 weeks. For co-cultivation analyses of breast cancer cells and adipose tissue explants, we set up a two-dimensional transwell system, which enables intercellular communication through soluble factors secreted into the medium but inhibits intermixture of the different cell types. Following 72 hours of co-culture with or without adipose tissue, total RNA was isolated from the breast cancer cells and subjected to microarray gene expression analyses.
