Time series of murine lungs after flu infection

Project description:Time course data from influenza A-infected mice were collected for up to 60 days after infection to monitor gene expression profiles of the host. Whole lungs of individual mice were harvested from three independent infection experiments and at least three biological replicates were taken for a single time point. Mock-infected mice served as controls. Early after infection, the activation of RIG-I and interferon pathways as well as the up-regulation of chemokine and cytokine expression revealed the innate immune response phase which was closely correlated with the presence of infectious virus. Subsequently, viral clearance was initiated by the onset of T cell infiltration. In the late phase of infection, the formation of tertiary lymphoid tissues, bronchus-associated lymphoid tissue (BALT), was observed. The formation of BALT resulted in permanent steady-state changes of the lung transcriptome.

Gene network analysis of interstitial macrophages after treatment with induced pluripotent stem cells secretome (iPSC-cm) in the bleomycin injured rat lung

Project description:Idiopathic pulmonary fibrosis (IPF) is a complex disease involving various cell types. Macrophages are essential in maintenance of physiological homeostasis, wound repair and fibrosis in the lung. Macrophages play a crucial role in repair and remodeling by altering their phenotype and secretory pattern in response to injury. The secretome of induced pluripotent stem cells (iPSC-cm) attenuates injury and fibrosis in bleomycin injured rat lungs. In the current study, we evaluate the effect of iPSC-cm on interstitial macrophage gene expression and phenotype in bleomycin injured rat lungs in vivo. Â iPSC-cm was intratracheally instilled 7 days after bleomycin induced lung injury and assessed 7 days later and single cell isolation was performed. Macrophages were FACS sorted and microarray analysis was performed. We characterized changes in the rat lung interstitial macrophages using transcriptional profiling.

Expression data from mouse luminal mammary epithelial cells

Project description:We used a mouse strain in which one Tbx3 gene was replaced with the yellow fluorescent protein variant Venus. Luminal cells had either very high Tbx3 promoter activity or not at all. We performed an expression analysis on luminal mammary epithelial cells sorted based on their Venus expression (reporting Tbx3 promoter activity) to investigate the difference between these two cell populations. Mammary epithelial cells from 3 Tbx3-Venus-KI adult virgin female mice (FVB background) were pooled and luminal cells were sorted into a Venus-hi and a Venus-neg sample. There were no repeats for this study.
