Transcription profiling by array of tumors excised from conditional mouse model of synovial sarcoma compared to skeletal muscle from wild type mice


ABSTRACT: The human SYT-SSX fusion protein was expressed in a developmentally dependent fashion in murine myoblasts. Tumors harvested from mice in adolescence were compared to normal mouse skeletal muscle samples. Experiment Overall Design: Murine tumors were excised from affected mice for RNA extraction and hybridization on Affymetrix microarrays. For control samples normal mouse skeletal muscle tissue was obtained and RNA extracted for hybridization to the Affymetrix arrays.

Synovial sarcoma is an aggressive soft-tissue malignancy marked by a unique t(X;18) translocation leading to expression of a chimeric SYT-SSX fusion protein. We report here a mouse model of synovial sarcoma based on conditional expression of the human SYT-SSX2. Using this model, we have identified myoblasts as a potential source of synovial sarcoma.
