Transcription profiling of human IOSE cells treated with TGFb1, time series

ABSTRACT: Unlike ovarian cancer, normal ovarian epithelium response to TGFb1 induced growth inhibition. This time course study tried to idenify genes that showed changes after additionof TGFb1 in immortalized ovarian surface epithelial cells (IOSE) which is derived from normal ovarian epithelial cells Experiment Overall Design: IOSE cells were treated with TGFb1 for 0, 3, 6, and 12hrs. Cells were harvested for total RNA extraction. Affymetrix expression microarray was performed to analyse gene expression changes.

Proteomics Deciphering the Underlying Mechanism of Tgfb1 Regulating Dendritic Cells

Project description:In current study, we conducted a lentivirus-based overexpression system and observed that overexpressing Tgfb1 successfully promoted the viability and altered the cell cycle distribution in DC. What’s more, this gene also greatly inhibited apoptosis of DC, highlighting the robust immune support effects of Tgfb1 function. Additionally, a quantitative proteomic was implemented to explore the underlying mechanism. Comparing Tgfb1 overexpressed (OE) and normal control (NC) groups, numerous differentially expressed proteins (DEPs) were emerged, which contributed to this phenomenon. Finally, bioinformatics analysis revealed multiple DEPs enriched in immune-related pathways directly or in a roundabout way, which manifested the crucial role played by Tgfb1 in DC.

Profiling fibrotic lung in TGFb1-Tg mice

Project description:Transgenic (Tg) mice over-expressing TGFb1 in a lung-specific manner display airway inflammation and fibrosis, phenotypes relevant to human COPD and asthma. This model system provides a platform for identifying and validating genes/pathways involved in these disease pathogeneses. The TGFb1-Tg mice lung profiling data was analyzed by identifying genes that were up- and down-regulated at selected p value and fold change in the lung of TGFb1-Tg mice compared to the corresponding wild type controls.

Effect of TGFb1 treatment and WISP1 overexpression on 4T1 breast cancer cells

Project description:This study set out to identify TGFb1 transcriptional targets in breast cancer cells. 4T1 breast cancer cells were treated with TGFb1 for 24h and transcripts that were significantly regulated were identified. Wisp1 was identified as one of the genes most highly upregulated by TGFb1. To identify the transcriptional targets of Wisp1, 4T1 cells were then virally transduced to increase Wisp1 expression. The gene expression profiles of Wisp1-overexpressing 4T1 cells were determined and revealed that Wisp1 overexpression does not result in major gene expression changes in 4T1 cells.

Each treatment was carried out in triplicate 10 cm plates and the experiment was carried out in triplicate using consecutive passages of cells.
