G72 overexpression in U87 cells

ABSTRACT: G72 is a susceptible gene for schizophrenia. No matter genetic variation or serum levels, G72 has been recognized as a biomarker for schizophrenia. In some reports, G72 genetic variation also related with bipolar disorder. Although G72 has been studied for 15 years, the protein function is still unclear. This study aims for predicting the biological function of G72 and hope to elucidate its biological role. two independent control (U87 cells transfected with empty vector) and two experiements (U87 cells transfected with G72 overexpression plasmid).

Increased plasma matrix metalloproteinase 1 is associated with the presence of esophageal squamous cell carcinoma

Project description:Esophageal squamous cell carcinoma (ESCC) patients are usually diagnosed at late stages, prohibiting the chance of surgical cure. The obstacle partly comes from the lack of molecular markers for early detection. Moreover, there is no consensus about endoscopic surveillance for ESCC. Identification of candidate targets for Haizao Yuhu Decoction acting on goiter rats

Project description:To predict the different genes, this study compared the gene expression level among the control group, the model group and the treatment groups. In this present study, 20 Wistar rats, weighting 160-180g and as the sex evenly, were randomly divided into five groups: normal control group, model control group , Haizao Yuhu decoction group, Haizao Yuhu decotion lacking Sargassum fusiforme (DL-S) group, and Haizao Yuhu decotion lacking Radix Glycyrrhiza (DL-G) group. Except the rats in control and model groups, the three other groups, which were established as goiter model, were administered appropriate medication at the dose of 1mL/100g for 28 consecutive days. After all animals sacrificed, thyroids were removed and then put in Trizol for microarray experiments.

The effect of vitamin D supplementation on tumorigenesis in a thioacetamide (TAA)-induced rat intrahepatic cholangiocarcinoma model

Project description:In this study, we investigated the effect of vitamin D supplementation on tumorigenesis in a thioacetamide (TAA)-induced rat intrahepatic cholangiocarcinoma model as vitamin D is known to have a spectrum of anticancer activities. Using PET, we found that tumor formation and progression were suppressed in rats fed a diet supplemented with 6 IU/g vitamin D3(+6D) as compared to the group fed a 2 IU/g vitamin D3 diet (+2D) or controls. Microarray analysis of the tumors that arose revealed that vitamin D supplementation caused significant up- and down regulation in 21 and 16 genes, respectively. There are 9 tissue samples (3 rats in each group labeled as (+6), (+2) and control). Duplicate analysis were used for each sample.

In the study, 50 male Kunming mice were randomly divided into 5 groups, namely, normal control group, model group, Daji/Gancao-synergy group, Daji/Gancao-antagonism group, Daji group. Normal control group and model group were intragastrically administrated with normal saline, while other 3 groups were intragastrically administrated with corresponding doses.

Identification of a biomarker panel for the intake of the common dietary trans fat elaidic acid (trans∆9-C18:1)

Project description:Elaidic acid is an abundant industrial produced trans fatty acid in foodstuffs. Trans fatty acid intake has been correlated to an unfavorable plasma lipoprotein profile and an increased cardiovascular disease risk. The present study aimed to identify a plasma protein biomarker panel related to human intake of elaidic acid. The human liver cell line HepG2-SF was used as a model system, and the cells were maintained for seven days in serum-free medium containing 100 µM elaidic acid (trans∆9-C18:1), oleic acid (cis∆9-C18:1) or stearic acid (C18:0). The secretomes were analyzed by stable isotope labeling of amino acids in cell culture (SILAC), difference in gel electrophoresis (DIGE) and gene expression microarray analysis. Twelve proteins were found to be differentially regulated based on SILAC data (>1.3 fold change, P-value < 0.05), 13 proteins were found to be differentially regulated based on DIGE analysis (>1.3 fold change, P-value < 0.05), and 17 mRNA transcripts encoding extracellular proteins were determined to be affected (>1.3 fold change, P-value < 0.01) following the addition of elaidic acid compared to oleic acid or stearic acid. The results revealed that 37 proteins were regulated specifically in response to elaidic acid exposure, and nine of these proteins were confirmed to be regulated in this manner by using selected reaction monitoring mass spectrometry. HepG2 cells kept used in serum free conditions. The cells were incubated for seven days in either 100 µM oleic, elaidic or stearic acid, before total RNA was extracted. For each group four biological replicates were made and from the each RNA extraction two technical replicates were made. There is a control group without fatty acid incubation.

Î±-catulin play a role in lung cancer metastasis

Project description:Background: Î±-catulin may functions as an oncoprotein, sustaining proliferation by preventing cellular senescence and promoting cancer cell migration. In this study, we investigated the mechanism of Î±-catulin in cancer cell migration and metastasis in lung cancer. Method: Î±-catulin mRNA expression was isolated from A549/AS2neo (control) and A549/AS2neo-Î±-catulin stable cells. The Phalanx Human OneArray microarray analysis was performed to identify Î±-catulin downstream genes. Results: Overexpression of Î±-catulin increased cancer cell migration and metastasis. By using Phalanx Human OneArray microarray we have identified panel of genes altered by Î±-catulin overexpression, consisting of CDC42, intergrins and genes related to cytoskeleton remodeling. Conclusion: Î±-catulin is an oncoprotein and promotes lung cancer cell migration and metastasis. Two-condition experiment, Vector vs. alpha-catulin overexpression cells.The cDNAs encoding full-length human alpha-catulin were amplified and subcloned into lentiviral pLKO_AS2.neo which generated full-length alpha-catulin. Vector control or alpha-catulin lentivirus were transduced into A549 cells and G418 was used to select stable cells.
