The role of PCNA ubiquitylation during unperturbed genome replication in S.pombe


ABSTRACT: To establish if PCNA ubiquitylation affects the DNA replication kinetics of specific loci, we compared rates of DNA replication across the genome between rad18+ and rad18-delta cells using the BrdU-IP sequencing method. We mapped BrdU-incorporation at 300-bp resolution and subsequent informatics analysis was performed to generate datasets of replication kinetics.

PCNA ubiquitylation on lysine 164 is required for DNA damage tolerance. In many organisms PCNA is also ubiquitylated in unchallenged S phase but the significance of this has not been established. Using Schizosaccharomyces pombe, we demonstrate that lysine 164 ubiquitylation of PCNA contributes to efficient DNA replication in the absence of DNA damage.
