Preadult life history variation determines adult transcriptome expression

ABSTRACT: Investigation of whole genome transcription expression level changes in Drosophila mojavensis wild-type populations (Las Bocas:LB and Punta Prieta:PP). The experiment was designed to investigate preadult life history affects over adult transcriptome expression. A total of 137 hybridizations were performed in this entire experiment. We used 135K 12-plex NimbleGen arrays. Total RNA was recovered from each sample listed below. The experimental design consisted a total of two populations (Las Bocas:LB; Punta Prieta:PP), two host diets (Agria:AG and Organ pipe:OP) and four "length of egg to adult development time" points, i.e. 14, 15, 16, and 17 days. (14 days:D1; 15 days:D2; 16 days:D3; 17 days:D4). Each chip measures the expression level of 14528 transcripts. One to 5 replicates were used for each type (R1, R2, R3 etc.). The experiment was designed to investigate life history transcriptomics in different environments. A total of 172 hybridizations were performed in this entire experiment. We used 135K 12-plex NimbleGen arrays. Total RNA was recovered from each sample listed below. The experimental design consisted a total of two populations (Las Bocas:LB; Punta Prieta:PP), two host diets (Agria:AG and Organ pipe:OP) and fourteen life stages (6 hr egg:E6; 1st instar larvae:L1; 2nd instar larvae:L2; 3rd instar larvae:L3; Early pupal stage:EP; Late pupal stage:LP; 0 Day old adult:0D; 3 Day old adult:3D; 4 Day old adult: 4D; 6 Day old adult:6D; 10 Day old adult:10D; 14 Day old adult:14D; 18 Day old adult:18D & 24 Day old adult:24D). Each chip measures the expression level of 14528 transcripts. Two to 5 replicates were used for each type (R1, R2, R3 etc.). Project description:Investigation of whole genome transcription expression level changes in Drosophila mojavensis wild-type populations (1 Punta Onah: PO, 2 Organ Pipe National Monument: OPNM, 3 Punta Prieta:PP, and 4 San Quintin: SQ). The experiment was designed to investigate functional genomic responses to temperature variation (15, 25, and 35 Â°C) in adult Drosophila mojavensis wild populations. For each treatment 1-5 replicates were used (R1, R2, R3, R4 & R5). SO and BC represents Sonora deserts and Baja California region respectively. A total of 97 hybridizations were performed in this entire experiment. We used 135K 12-plex NimbleGen arrays. Total RNA was recovered from each sample listed below. The experimental design consisted a total of four populations (Punta Onah:PO; Organ Pipe National Manument:OPNM, Punta Prieta:PP and San Quintin:SQ), two host diets (Agria:AG and Organ pipe:OP) and three temperature treatments (15, 25 and 35 Â°C). Each chip measures the expression level of 14528 transcripts. One to 5 replicates were used for each type (R1, R2, R3, R4 and R5). Project description:Investigation of whole genome transcription expression level changes in Drosophila mojavensis wild-type populations (1. Punta Onah:PO; 2. Organ Pipe National Manument:OPNM; 3. Punta Prieta:PP; & 4. San Quintin:SQ). The experiment was designed to investigate effects of desiccation exposure (0, 9 & 18 hr) and host plant (diet) on transcriptome. A total of 95 hybridizations were performed in this entire experiment. We used 135K 12-plex NimbleGen arrays. Total RNA was recovered from each sample listed below. The experimental design consisted a total of four populations (1. Punta Onah:PO; 2. Organ Pipe National Manument:OPNM; 3. Punta Prieta:PP; & 4. San Quintin:SQ), two host diets (Agria and Organ pipe) and three desiccation treatments (0, 9 and 18 hours). Each chip measures the expression level of 15528 transcripts. The experiment was designed to investigate effects of host plant (diet), mating status (mated:M or nonmated:V) and sex (male:male or Female:F) on transcriptome. A total of 128 hybridizations were performed in this entire experiment (127 in the final analysis). We used 135K 12-plex NimbleGen arrays. Total RNA was recovered from each sample listed below. The experimental design consisted a total of four populations (1. Punta Onah:PO; 2. Organ Pipe National Monument:OPNM; 3. Punta Prieta:PP; & 4. San Quintin:SQ), two mating status (mated:M or nonmated:V), both the sexes (male:male or Female:F) fed on two different diet regimes (Agria and Organ pipe). Characterization of pain-related behaviors and gene expression profiling of peripheral sensory ganglia of a mouse model of acute ankle sprain

Project description:Lateral ankle sprain (LAS) is a very common type of joint injury.Unfortunately, a certain proportion of individuals who once developed LAS may suffer persistent ankle pain and recurrent sprains. However, the molecular pain mechanisms underlying LAS are still not well understood. To investigate the potential mechanisms of pain in LAS model mice, we performed RNA- Seq of ipsilateral DRG of sham and LAS model group mice. GO analysis found that the mostly enriched biological process include immune response and positive regulation of gene expression. Immune response can trigger neuroinflammation, a critical process for peripheral sensitization and contributes to chronic pain mechanisms. Furthermore, our RNA-Seq analysis indicated that LAS modulated the expression of multiple genes and gene networks in the DRGs. Our work may help to further advance the understandings of the molecular pain mechanisms underlying LAS and help to identify novel targets for LAS.
