X chromosome cDNA microarray screen of lymphoblasts from males with XLMR

ABSTRACT: The goal of this study is to identify genes that show altered transcript levels in lymphoblasts from male patients with XLMR as candidate genes that are responsible for XLMR. Keywords: disease state Using a custom X chromosome cDNA microarray, we studied X chromosome gene expression in lymphoblasts from 43 patients with XLMR and identified those genes with altered transcript levels as compared to that in 6 normal controls.

X-linked Mental Retardation (XLMR) occurs in 1 in 600 males and is highly genetically heterogeneous. We used a novel human X chromosome cDNA microarray (XCA) to survey the expression profile of X-linked genes in lymphoblasts of XLMR males. Genes with altered expression verified by Northern blot and/or quantitative PCR were considered candidates. To validate this approach, we documented the expected changes of expression in samples from a patient with a known X chromosome microdeletion and from p  ...[more]
