Gene expression of FACC canine cancer cell line panel (Affymetrix Canine 2.0 array)

ABSTRACT: Cancer cell line panels that incorporate genomic and pharmacologic screening data have become powerful tools for investigating the relationship between gene expression and drug response in human cancer research. Similar cell line panels of such a scale as the NCI60, CCLE, or GDSC do not currently exist in canine cancer research. Here we have assembled a canine cancer cell line panel of 29 cell lines representing multiple tumor types that have undergone gene expression profiling and have begun to be used for pharmacologic screening. RNA was extracted from drug naÃ¯ve canine cancer cell lines and subjected to microarray analysis.

Project description:The Cancer Cell Line Encyclopedia (CCLE) project is a collaboration between the Broad Institute, the Novartis Institutes for Biomedical Research and the Genomics Novartis Foundation to conduct a detailed genetic and pharmacologic characterization of a large panel of human cancer models It consists of a compilation of gene expression, chromosomal copy number, and massively parallel sequencing data from nearly 1,000 human cancer cell lines. All raw and processed data are available through an integrated portal on www.broadinstitute.org/ccle The final cell line collection spans 36 cancer types. Representation of cell lines for each cancer type was mainly driven by cancer mortality in the United States, as a surrogate of unmet medical need, as well as availability.

miRNA expression of canine mammary cancer stem cells

Project description:The aim of our study was to assess miRNA expression of canine mammary cancer stem cells. Three canine mammary cancer cell lines (CMT-U27, CMT-U309 and P114) were stained using Anti-Sca1 (Stem cell antigen 1) antibodies. The FACS analysis showed 0,3-1% of Sca1+ cells in each of the cell line. The cells were sorted (using FACS Aria II) as Sca1+ and Sca1- and subjected to further analysis of miRNA expression (using Agilent custom miRNA microarray).

Global gene expression of canine bladder K9TCC cells and cancer stem cells after COX-2 inhibition

Project description:Cancer stem cells (CSCs) are fundamental to bladder cancer progression and the cyclooxygenase-2 (COX-2) pathway has a pro-tumorigenic role in CSC function. Naturally occurring bladder cancer in dogs has the potential to be a natural model of the human disease. Here, we used canine specific microarrays to determine changes in global gene expression of canine bladder cancer cells (the K9TCC cell line) and bladder cancer stem cells isolated from the K9TCC cell line after treatment with the selective COX-2 inhibitor mavacoxib or a COX-2 siRNA.
