Project description:The transcriptional response of H. armigera larvae was analyzed after feeding on gossypol supplemented diet, a toxic secondary metabolite produced in cotton plants, to detect potential detoxification enzymes possibly involved in detoxification of gossypol by H. armigera. A one-color microarray-based gene expression analysis was performed with Cy3 labeled cRNA of gut and rest of the body of H. armigera larvae that fed on 0.016%, 0.16% gossypol and control diet. For each treatment and tissue four biological replicates were used.

Project description:Transcriptional profiling comparing gut tissue from fifth instar larvae exposed to six different diets, namely cotton fruit or boll (B), tobacco flower bud (TFB), bean pod (BP), chick pea fruit (CKPF), pinto bean-based artificial diet (PB) and wheat-based artificial Lepidoptera diet (BIO). The generalist lepidopteran herbivore Helicoverpa armigera can feed on more than 87 plant species belonging to 48 families. However, life table studies on different crops have revealed that cotton and corn are the most suitable hosts of this pest and tomato, hot pepper and tobacco are suboptimal. It is believed that generalists owe their success to the deployment of various members of multigene families of detoxicative and digestive enzymes, a strategy that may also be responsible for rapid evolution of insecticide resistance. However studies of generalist adaptations have been limited to specific genes or gene families, and an overview of how these adaptations are orchestrated at the transcriptional level is lacking. We compared the transcript profiles of larval guts in response to differentially suitable hosts and towards two different artificial diets commonly used for laboratory rearing of this species, using a two-color alternating loop design microarray experiment. Project description:The tobacco hornworm, Manduca sexta, is a lepidopteran model species widely used to study insect biochemical processes. While some of its larval hemolymph proteins are well understood, a detailed proteomic analysis was unavailable until 2016, revealing features such as correlation with transcriptome data, formation of immune complexes, and constitution of an immune signaling system. Yet, it is unclear how these may change in other developmental stages. In this paper, we report the proteomes of cell-free hemolymph from prepupae, pupae on days 4 and 13, and young adults. Of the 1,824 proteins identified, 907 have a signal peptide and 215 are related to immunity. Drastic changes in abundance of the storage proteins, for instance, reflect physiological disparities among prepupae, pupae, and adults. Considerably more proteins lacking signal peptide are present in the late pupae, suggesting that plasma acts as a temporary reservoir for intracellular components released from remodeling tissues during metamorphosis. In summary, the proteins and their levels revealed in this study are expected to stimulate focused explorations of humoral immunity in wandering larvae, pupae, and adults of M. sexta and shed light upon functional and comparative genomic research in other holometabolous insects.

Project description:Aphid adaptation to harsh winter conditions is illustrated by an alternation of their reproductive mode. Aphids detect photoperiod shortening by sensing the length of the night and switch from viviparous parthenogenesis in spring and summer, to oviparous sexual reproduction in autumn. The photoperiodic signal is transduced from the head to the reproductive tract to change the fate of the future oocytes from mitotic diploid embryogenesis to haploid formation of gametes. Because of viviparous parthenogenesis, the whole process takes place in three consecutive generations. To understand the molecular basis of the switch in the reproductive mode, a transcriptomic approach was used to detect significantly regulated transcripts in the heads of the pea aphid Acyrthosiphon pisum. The transcriptomic profiles of the heads of the first generation were slightly affected by photoperiod shortening. This suggests that trans-generation signaling does not occur between the grand-mothers and the viviparous embryos they contain. By analogy, many of the genes regulated in the heads of the second generation are implicated in visual functions, photoreception and cuticle structure. The modification of the cuticle could decrease the storage of N-β-alanyldopamine and provoke an increase in free dopamine concentration. Based in results in Drosophila, modification of the insulin pathway could cause a decrease of juvenile hormones in short-day reared aphids. Biological material for microarray experiments was prepared under two dayly photoperiodic regimes both at constant temperature of 18°C: i) âShort Nightâ (SN) at 16h of light and ii) âLong Nightâ (LN) at 12h of light to induce the production of sexual morphs. To initiate the experiment, two groups of 105 L3 larvae were placed either under SN or LN condition. This corresponds to generation G0. At the middle of the photophase, 25 individual were frozen when they had reached both the L4 and the wingless adult (WA) stages, in the two photoperiod conditions. The 55 remaining WA individuals (still divided in two groups) were left on 55 plants to lay their offspring: one larva of the 1st stage (L1) was kept per WA. This larva was selected among the 20 first born larvae. This is the generation G1. At the middle of the photophase, 25 individual were frozen when they had reached both the L2 and the L4 stages, in the two photoperiodic conditions. Thus, 25 individuals from 4 different stages (L4-G0, WA-G0, L2-G1 and L4-G1) were collected in the two photoperiod conditions with 3 biological replicates, forming the 24 samples used for microarray experiments. RNAs from heads of aphids from the two photoperiodic conditions were hybridized one against the other for each stage with a dye-swap.The experimental design is thus 24 arrays which corresponds to the described samples of that series.

Project description:Gut and rest of body tissue from fifth instar larvae which fed for three days a diet containing different doses of gossypol. Transcriptional profiling comparing gut and rest of body samples for three gossypol concentrations (0%, 0.016%-hormetic dose & 0.16%-detrimental dose). Hormesis is a biphasic biological response characterized by the stimulatory effect at relatively low amounts of chemical compounds otherwise known to be detrimental at higher concentrations. A hormetic response in larval growth rates has been observed in cotton-feeding insects in response to increasing concentrations of gossypol, a toxic metabolite found in the pigment glands of some Malvaceae plants. We investigated the developmental effect of gossypol in the cotton bollworm, Helicoverpa armigera, an important heliothine pest species, by exposing larvae to different doses of this metabolite in their diet. In addition, we sought to determine the underlying transcriptional responses to different gossypol doses. Larval weight gain, pupal weight and larval development time were measured in feeding experiments and a hormetic response was seen for the first two characters. On the basis of net larval weight gain responses to gossypol, three treatments, that is 0%, 0.016% and 0.16% gossypol , were selected for transcript profiling in the gut and the rest of the body in a two-color double reference design microarray experiment. Project description:To allow estimation of the complexity of the antennal transcriptome between sexes as well as in comparison with a non-antennal tissue, microarrays were designed based on an assembly of new 454 sequence data from the antenna and publicly available data including 454 data from the larval midgut. The microarrays were hybridized with samples generated from the repsecitve tissues from 3 (antenna) and 5 (midgut) animals per sample, with four independent samples per sex (antenna) and tissue (midgut). Sequence assembly of included Manduca antennal and gut ESTs (63) and all publicly available Genbank sequences was used with eArray (Agilent Technologies) for the design of 4 x 44K microarrays based on 60mer oligo probes. For sex-specific antennal microarray hybridizations, RNA of three individuals of one sex was pooled per preparation, and 5 larvae each were dissected for gut tissue isolation with four biological replicates per sex (antennae) and tissue (gut), respectively. Double purified total RNA was added to Agilent Technologies spike-in RNA and labeld using QuickAmp Amplification kit (Agilent Technologies) and the Kreatech ULS Fluorescent Labeling Kit with cyanine 3-CTP dye following the manufacturer´s instructions. Amplified cRNA samples were used for microarray hybridization, scanned with the Agilent Microarray Scanner and data was extracted from TIFF images with Agilent Feature Extraction software version 9.1. Raw data output files were analyzed using the GeneSpring GX11 and GeneSifter microarray analysis softwares.
