Analysis of gene expression in hypoxic and normoxic HeLa cells.

ABSTRACT: The purpose of experiment is to compare the mRNA expression differences between HeLa cells conditioned with normoxia 21% oxygen and hypoxia 1% oxygen at 6h and 20h. Total RNA was obtained from HeLa cells for 4 different conditions, 3 biological replicate experiments were performed, resulting in a total of 12 samples.

Project description:Human immature dendritic cells derived from monocytes were activated to become mature dendritic cells by exposure to platebound Collagen I or Collagen. The importance of OSCAR-signalling was evaluated by pre-incubating the immature dendritic cells with an inhibitory monoclonal antibody to the OSCAR receptor, or an isotype control. As a positive control, immature dendritic cells were activated by using LPS. The set-up was examined at 4 hours and 20 hours to evaluate early and late effects of OSCAR signalling.

Genome-wide analysis of gene expression perturbed by reptin shRNA in MCF7 subjected to normoxic and hypoxic conditions

Project description:In order to investigate the role of reptin methylation on the expression of hypoxia-responsive genes across the whole genome, we performed a microarray analysis from RNAs isolated from MCF7 cells expressing either control shRNA (shRNA) or reptin shRNA (shreptin) in normoxic and hypoxic conditions. MCF7 cells were transiently transfected with either non-specific shRNA or reptin shRNA in replicates. Cells were grown for 24 hours before being exposed to either normoxic (21% oxygen) or hypoxic (1% oxygen) conditions for 18 hours. Cells were then lysed and RNA was isolated.

Comparison of receptor tyrosine kinase (RTK) family gene expression between normoxic cells and hypoxia-induced gefitinib resistant clones

Project description:In this study, we explored the mechanisms of hypoxia-induced EGFR TKI resistance in non-small cell lung cancer (NSCLC) harbored activating EGFR mutation. The NSCLC cell lines were exposed to normorxia or 1% oxygen for 4 weeks, and then we tested EGFR TKI sensitivity in normoxic and hypoxic NSCLC cell lines. In this microarray experiment, we used normoxic HCC827 and hypoxia-induced gefitinib resistant clones, C2-3 and C2-10. Those clones were selected with gefitinib treatment after the HCC827 were exposed to 1% oxygen for 4 weeks, and the HCC827 C2-3 and C2-10 clones were selected at random for this study.
