Differential expression data for clonal EL4 mouse lymphoma cell lines

ABSTRACT: Our group originally developed clonal cell lines by limiting dilution of the parental cell line, and also stably transfected one of these cell lines with human phospholipase D2. The expression analysis was done to examine differences between a non-metastatic cell line (WT2), a metastatic cell line (V&), and a highly metastatic cell line (C5) that had been transfected with hPLD2. The cell lines, which had been serum-starved overnight, were analyzed without further treatment.

Project description:Three metastatic melanoma cell lines (BD-0548-ME, DP-0574-ME and WP-0614-ME) were transfected with miR precursors: hsa-miR negative control (NC) or hsa-miR-200a-3p precursors (miR-200a). The objective of this experiment was to determine miR-200a targets in metastatic melanoma cell lines. We selected low -miR-200a expression cell lines and then we overexpressed miR-200a or NC. Finally, we compared the metastatic melanoma cell lines with miR-200a overexpression (miR-200a) to the same cell line transfected with the negative control miR (NC).

Transfection of rat Sertoli cells with human androgen receptor leads to alternated gene expression without androgen incubation

Project description:We introduced a full-length human AR into the expression vector pcDNA 6.2 C-EmGFP and transfected AR-deficient rat Sertoli cells (93RS2) by electroporation. We compared gene expression levels of AR-deficient Sertoli cell line (93RS2_noAR) and a full-length-human-AR transfected Sertoli cell line (93RS2_AR17) using microarray analysis. We analyzed 3 arrays for the noAR cell line and 3 arrays for the AR17 cell line.

A Novel Orthotopic Mouse Model of Head and Neck Cancer and Lymph Node Metastasis

Project description:Analysis of the genes involved in the metastasis of head and neck squamous cell carcinoma (HNSCC). We hypothesized that there would be overexpression of certain genes in the highly metastatic USC-HN1-GFP-G1 and USC-HN1-GFP-G2 cell lines compared to the parental USC-HN1-GFP cell line. We used the USC-HN1 cell line stably transfected with H2B-GFP and developed an immortal cell line (USC-HN1-GFP), which was injected into the tongues of SCID/nude mice. The metastatic cells were collected at the LN, separated and cultured, forming an immortal USC-HN1-GFP-G1 cell line. These cells were reinjected into the tongues of mice and the USC-HN1-GFP-G2 cell line was formed by the same LN collection method. The gene expression in these three cell lines were compared using Illumina microarray chips.

Human melanoma cell lines secretome analysis

Project description:The imbalance of cellular homeostasis during oncogenesis together with the high heterogeneity of tumor-associated stromal cells have a marked effect on the repertoire of the proteins secreted by malignant cells (the secretome). Hence, the study of tumoral secretomes provides insights for understanding the cross-talk between cells within the tumor microenvironment as well as the key effectors for the establishment of the pre-metastatic niche in distant tumor sites. In this context, we performed a proteomic analysis of the secretomes derived from four cell lines: (i) a paired set of fibroblasts - Hs 895. T, a cell line obtained from a lung node metastatic site from a patient who had melanoma and Hs 895.Sk, a skin fibroblast cell line (derived from the same patient); (ii) two malignant metastatic melanoma cell lines - A375, a malignant melanoma cell line from primary source and SH-4, a cell line derived from pleural effusion of a patient with metastatic melanoma. Clustering of expression profiles together with functional enrichment revealed patterns that mirrored each cell type (skin fibroblasts, cancer-associated fibroblasts and metastatic cells). These patterns might be the result of cell-specific protein expression programs and may serve as basis for further proteomic analysis of melanoma cell lines secretomes.

Transcription profiling by array of a PaTu-8988s and PaTu-8988t cell line differentiation model

Project description:PaTu-8988s and PaTu-8988t are two cell lines which were derived from the same liver metastasis of a human primary pancreatic adenocarcinoma, but differ in their differentiation status and metastatic capacity. The poorly differentiated cell line PaTu-8988t was transfected with an E-Cadherin expression construct or vector control, respectively, and individual clones analyzed by expression profiling. In addition, 3 independent preparations of wild-type PaTu-8988s cells were analyzed.
