Each cell line (both parental and resistant) were then treated with DMSO (control), capivasertib monotherapy, fulvestrant monotherapy and capivasertib/fulvestrant combination. RNA data is available after each treatment and resistant cell lines additionally have RNA data available after continuous palbociclib treatment.

Project description:Oral selective estrogen receptor degraders (SERDs) could become the backbone endocrine therapy (ET) for estrogen receptor-positive (ER+) breast cancer, as they achieve greater inhibition of ER-driven cancers than current ETs and overcome key resistance mechanisms. This dataset was aimed at exploring the preclinical effects of the next-generation oral SERD camizestrant (AZD9833) on MCF7 and CAMA1 cell lines. Cells were treated with 1 nM estradiol and 100 nM fulvestrant, AZD9496, or camizestrant for 24 hours, with three replicates per condition and non-estradiol-treated controls.

Project description:This experiment aimed at identifying transcriptome-wide differences in a murine model of epilepsia after treatment with soticlestat. We used three different cell populations (neurons, microglia, and astrocytes) from soticlestat-treated mice, mice where the epileptic state had been induced, and sham mice as control. We compared the differences within each group (treatment / status epilepticus versus sham) and across groups (treatment - status epilepticus) to identify genes involved in the disease, and on the other hand, genes involved in the therapeutic effect of the drug.

Project description:Analysis of transcription response of undifferentiated human BE(2)-C neuronal cells to stimulation with purified antimycin A1a or unfractionated commercially available antimycin A (Sigma A8674). Three independent sets of BE(2)-C cells were treated DMSO control or 200 nM of either purified antimycin A1a or unfractionated antimycin A. Total RNA was harvested at 24 h after stimulation and analyzed with Affymetrix U133 Plus 2.0 chips.

Project description:As supplies of monocytes, macrophages and dendritic cells from human sources can be scarce or prone to donor variation we established an efficient method to generate induced pluripotent stem cell derived monocytes that in turn could be differentiated into both macrophages and dendritic cells. We used RNA sequencing to profile these from multiple differentiation runs (n=3) and multiple monocyte harvests (n=3-4) and compared them to their blood derived counterparts, blood derived monocyte, monocyte derived macrophages and moncyte derived dendritic cells (from 3 donors).
