Transcription profiling of liver from rats treated with triiodothyronine at 0.1ug or 5ug after 1 or 3 hours

ABSTRACT: A single dose of 5 ug of Triiodothyronine (T3) was delivered subcutaneously to adult rats to stimulate hepatocyte proliferation. This was compared to rats treated without T3, or with a a submitogenic dose (0.1 ug). Livers were isolated and harvested at 1 h and 3 h post administration. A comparison was performed between no T 3 and 5 ug of T3 at 3 h and between 5ug T3 and 0.1 ug of T3 at 1 h. RNA was run on Affymetrix RAE230+2.0 arrays.

Background and aim
A single dose of the thyroid hormone tri-iodothyronine, T3, can enhance both size and function of normal rodent liver, which is potentially of value in the treatment of liver disease. However the mechanism of this has not been fully elucidated, and it cannot be modeled in vitro. We therefore investigated the transcriptome response to T3 in rat liver in vivo.
Methods
After adult rats were administered 5 microg T3 subcutaneously, a whole rat genome microarray comp ...[more]
