Transcription profiling of Lotus japonicus major organ systems including nodule and seed development

ABSTRACT: Lotus japonicus is a perennial legume with a small diploid genome that has been adopted as a model species for legume genetics and genomics. With the genome sequence as a backdrop (Sato et al. 2008), we have generated a gene expression atlas that provides a global view of gene expression in all major organ systems of this species, including nodule and seed development.

Transcrition profiling of shoots from related model and cultivated Lotus plant species following salt acclimation

Project description:Many plant researchers have applied genomic tools to model species to identify abiotic stress responsive genes that might be useful for improving stress tolerance in crops. However, it is unclear whether this translational approach will be successful given the complexity of abiotic stress tolerance. We carried out a functional genomic (ionomic, transcriptomic and metabolomic) comparison of three model and three forage species of the genus Lotus with varying tolerance to salinity. Transcriptome analysis showed that about 60 % of expressed genes were responsive to salt treatment in one or more of the six species tested, but less than 1 % was responsive in all genotypes. Therefore, genotype-specific responses overshadowed conserved transcriptional responses to salinity and represent an impediment to translational genomics. Fortunately, 'triangulation' from multiple species enabled the identification of a core set of conserved and tolerant-specific responses that could provide durable tolerance across genotypes.

Genome wide analysis of glucose brassinosteroid interaction in Arabidopsis

Project description:Arabidopsis thaliana seeds after imbibition were inoculated in ½ MS medium supplemented with 0.8% agar and 1% sucrose. Once the plant material was uniformly germinated, the experimental conditions were applied. 5d old light-grown uniformly germinated seedlings were washed seven times with sterile water with last wash given by ½ MS liquid medium without sucrose to remove residual exogenous sugar and the plant material was kept in ½ MS liquid without sucrose in the dark for all subsequent steps. Cultures were shaken at 140 rpm at 22oC for 24 h and then 3 h treatment was given with liquid ½ MS without glucose and liquid ½ MS supplemented with BR (0.1 ?M EBR), glucose (3%), glucose (3%) + BR (0.1 ?M EBR). Seedlings were harvested after 3h and preceded for RNA isolation and Microarray analysis.
