Transcription profiling of human osteosarcoma Sa OS cells vs reference RNA of human osteosarcoma U-2 OS cells

ABSTRACT: Osteosarcoma (OS) is a common primary bone malignancy that is characterized by high degree of aneuploidy, gene amplification, and multiple unbalanced chromosomal rearrangements. The human osteosarcoma U-2 OS and Sa OS cells lines have been generated more than three decades ago and are used in a wide spectrum of biomedical research. Nevertheless, and despite scattered information about their genetic context, no comprehensive comparative study of their transcriptome profile has been reported to date. The aim of this study was to elucidate common molecular characteristics of the two cell lines as well as differences in their expression profile. Thus the genome wide gene expression profile of the Sa OS cells was compared with reference RNA of U-2 OS cells. These results may provide the basis for future studies and illuminate the molecular differences of the two widely used cell lines.

Changes in gene expression in HOS and U2-OS cell lines after exposure to 600 Î¼g/mL OGE

Project description:Osteosarcoma (Osteosarcoma) is a type of bone cancer. Eighty percent of this tumor will be metastatic to the lungs or liver, and as a result, patients generally need chemotherapy to improve survival possibility. Recently, anti-tumor activity has been reported in Ocimum gratissimum aqueous extract (OGE), which has been the focus of recent extensive studies on therapeutic strategies due to its antioxidant properties. We used microarrays to identify potential and novel target genes responsive to the anticancer effect in OGE treatment in osteosarcoma cells, We performed pharmacogenomics analyses for the effect of OGE on human osteosarcoma U2-OS and HOS cell growth. Cell viability, Western blot and flow cytometry analysis were performed before performing pharmacogenomics analyses for the effect of OGE on human osteosarcoma U2-OS and HOS cell growth, including cDNA microarray and RT-PCR assays.

Project description:Four cultures (four biological replicates) of each of two strains, wild type H37Rv and a deletion mutant for mce3R gene, were grown in in Middlebrook 7H9 medium supplemented with 0.05% Tween 80, or Middlebrook 7H11, supplemented with albumin 0.5%, dextrose 0.4%, and 0.5% glycerol (M7H9-AD-G). <br>Total RNA was extracted from each culture, dyed with Cy5 and hybridised onto two separated microarrays (technical duplicates). <br>In the second channel a mix of genomic DNA from strains H37Rv and AF2122 was dyed with CY3 and used as a common control in all the microarrays.<br>An statistical analysis of microarray data was performed to detect genes with differential expression between genotypes.

Project description:Proteomic experiments were performed on the outer segment (OS) and the remaining retina (RR). For the OS proteomics, three biological replicates were performed for mouse at the age of P15, which included 24, 26 and 26 retinas respectively. We also performed the OS proteomic experiment on 40 retinas from mice at age P13. For the RR proteomic experiments, two biological replicates were performed independently using 30 and 30 retinas respectively.
