Transcription profiling of livers of adult UPF2 null mice in response to partial hepatechtomy

ABSTRACT: Deletion of the NMD component UPF2 in adult liver and response to partial hepatechtomy. Lethally irradiated Upf2flox/flox and Upf2flox/flox; Mx1Cre animals were transplanted with WT BM at 10-12 weeks of age. 5-6 following transplantion the bone marrow transplanted (BMT) animals were injected 3 times with poly-IC (Days 0, 2, 4), which facilitates Cre-mediated deletion in BMT Upf2flox/flox; Mx1Cre (hitherto UPF2 null) but not in BMT Upf2flox/flox control (hitherto WT) mice. A day 14 mice were subjected to partial hepatechtomy (0 hours) and sacrificed after 36 hours (36 hours)..

Background
Nonsense-mediated mRNA decay (NMD) is a post-transcriptional RNA surveillance process that facilitates the recognition and destruction of mRNAs bearing premature terminations codons (PTCs). Such PTC-containing (PTC+) mRNAs may arise from different processes, including erroneous processing and expression of pseudogenes, but also from more regulated events such as alternative splicing coupled NMD (AS-NMD). Thus, the NMD pathway serves both as a silencer of genomic noise and a re ...[more]
