Transcription profiling of human fibrotic liver samples

ABSTRACT: Human liver samples were obtained from Analytical Biological Services Inc.(Wilmington, DE, USA) via autopsy 4-10 hours post-mortem with appropriate consent. The specimens were quickly frozen and stored at -80C. Following pulverization of the liver specimens total RNA was isolated by QIAzol extraction and RNeasy Mini Kit(QIAGEN, Valencia, CA, USA) purification. The RNA was then amplified via Ambion
5X MessageAmp II aRNA Amplification Kit (Austin, TX, USA) and profiled using Agilent Human Whole Genome Microarray Kit (Agilent Technologies, Santa Clara, CA, USA).

ORGANISM(S): Homo sapiens

Unlabelled

Background
The possibility of extracting RNA and measuring RNA expression from paraffin sections can allow extensive investigations on stored paraffin samples obtained from diseased livers and could help with studies of the natural history of liver fibrosis and inflammation, and in particular, correlate basic mechanisms to clinical outcomes.
Results
To address this issue, a pilot study of multiplex gene expression using branched-chain DNA technology was conducte ...[more]
