Transcriptomics of drougth stress in L. japonicus under normal air conditions

Project description:L. japonicus (Regel) Larsen cv, Gifu were initially obtained from Prof. Jens Stougaard (University Aarhus, Denmark) and then self-propagated at the University of Seville. Seeds were scarified and surface-sterilized, germinated in 1% agar Petri dishes, and transferred to pots using a 1:1 (v/v) mixture of vermiculite and sand as solid support. Five seedlings were planted in each pot and grown during 35 days in a growth chamber under 16/8 hours day/night, 20/18M-oM-?M-=C, with a photosynthetic photon flux density of 250 M-oM-?M-=mol/m2M-oM-?M-=s and a constant humidity of 70%. Plants were watered with Hornum nutrient solution. Drought was applied withholding irrigation for the reported period of time and sample plants or leaves were harvested for further analysis.

Transcription profiling of Listeria monocytogenes after infection into murine macrophages

Project description:P388D1 murine macrophages were cultured in 85 mm tissue culture plates to about semi confluency. L. monocytogenes serotypes (1/2a EGD-e, 4a L99, 4b CLIP80459 and 4b F2365) were infected to the P388D1 cell monolayer at a MOI of 100 per eukaryotic cell. Infection was carried for 45 min and followed by addition of fresh medium containing 20 M-5g/ml gentamicin. The medium of the plates (containing 20 M-5g/ml gentamicin) infected with L. monocytogenes serotypes were replaced after 2 h post infection with fresh medium containing 50 M-5g/ml gentamicin. At each step the plates were washed extensively with 1x PBS. Incubation of the bacterial tissue culture plates was carried out in a humidified incubator for up to 4 h post infection.

Global transcriptional changes of Clostridium acetobutylicum cultures with increased butanol-acetone ratios

Project description:Artificial electron carriers have been widely used to shift the solvent ratio towards butanol in acetone-butanol-ethanol (ABE) fermentation of solventogenic clostridia according to decreased hydrogen production. In this study, first insights on the molecular level were gained to explore the effect of methyl viologen addition to cultures of Clostridium acetobutylicum. Employing batch fermentation in mineral salts medium, the butanol:acetone ratio was successively increased from 2.3 to 12.4 on a 100 ml scale in serum bottles and from 1.4 to 16.5 on a 1,300 ml scale in bioreactors, respectively. The latter cultures were used for DNA microarray analyses to provide new information on the transcriptional changes referring to methyl viologen exposure and thus, exhibing gene expression patterns according to the manipulation of the cellular redox balance.

Microarray-based transcription profiling of Streptococcus pneumoniae R6 delta rgtA ,delta rgtD and delta cpoA

Project description:Alterations in genes for penicillin-binding proteins (pbp) are well-known determinants for the resistance of Streptococcus pneumoniae to B-lactam antibiotics. Surprisingly, some mutations in non-pbp genes were also found to contribute to B-lactam resistance. Two of them discovered in the piperacillin resistant mutants P106 and P104, affect the expression of cpoA (encoding a glycosyltransferase) and of the rgtABCDHR cluster (encoding two small membrane proteins, an ABC transporter and a regulatory two-component system), respectively. cpoA and rgtABCDHR are involved in maintaining the synthesis and the proper ratio of the two major membrane glycolipids, and deletions in these genes led to complex phenotypes. In attempts to identify genetic determinants for these phenotypes, the global trancription patterns of the deletion mutants R6 delta cpoA, R6 delta rgtA and R6 delta rgtD were compared to that of the parent strain R6.
