Transcription profiling by array of human peripheral blood lymphocytes or haemopoietic stem cells at the time of transduction with a gammaretroviral vector

ABSTRACT: Expression profile comparison of peripheral blood lymphocytes (PBL) or haemopoietic stem cells (HSC) at the time of transduction with a gammaretroviral vector. The analysis is performed in the context of two different gene therapy clinical trials (Recchia et al, 2006; Aiuti et al, 2007) based on autologous infusion of human T cells or CD34+ haemopoietic progenitor cells gene-corrected with an moloney leukemia virus-derived vector (MLV-vector). T cells and CD34+ cells were isolated from patients and pre-stimulated in vitro with cytokines prior to transduction with the vector and re-infusion in patient. The expression profiles of T cells and CD34+ cells were analysed by microarray just after cytokine stimulation, providing information on the transcriptional activity of these two target cells at the time of transduction.

Project description:Different fusion oncogenes in acute myeloid leukemia (AML) have distinct clinical and laboratory features suggesting different modes of malignant transformation. Here we compare the in vitro effects of representatives of major groups of AML fusion oncogenes on primary human CD34+ cells. For the 3 d and 8 d time points, the oncogenes were retrovirally expressed in primary human CD34+ cells, with cells transduced with empty retroviral vector as controls. As retroviral transduction requires days, for the 6 h time point the oncogenes were expressed from a pTracer-CMV/Bsd plasmid using nucleofection. Cells nucleofected with empty pTracer-CMV/Bsd vector were used as a control.

Understanding hematopoietic stem cell mobilization and engraftment

Project description:This study looks at patients undergoing gene therapy for genetic blood and immune disorders, in particular sickle cell disease. From each patient, single blood HSPC colonies are grown from patient blood mononuclear cells, bone marrow mononuclear cells, or CD34+ at multiple time points: pre gene transduction, post-transduction, and post-infusion (at several time points). Whole-genome sequencing will be performed on DNA from these colonies and somatically-acquired mutations will be called. Using these mutations, a phylogeny of these cells can be inferred and further inferences can be made about the gene therapy transduction and engraftment steps.
