Transcription profiling of clofibrate treated rat livers


ABSTRACT: 5 day q.d. treatment by oral gavage of SD rats using 0, 200, 400 or 800 mg/kg/day of the peroxisome proliferator clofibrate. Livers were removed, RNA was isolated, and Cy3/Cy5 probes generated for 2-channel microarray experiments.

Peroxisome proliferators such as the fibrates act via the peroxisome proliferator activated receptor (PPAR)-alpha as hypolipidemic agents. Many peroxisome proliferators are also nongenotoxic hepatic carcinogens and hepatotoxicants in rodents. We performed transcription profiling using cDNA microarrays on livers of rats treated for 5 days with 3 doses of the peroxisome proliferator clofibrate.
