Project description:We found that the antibiotic colistin acts synergistically with antifungals of the echinocandin class (e.g. aminocandin) on C. albicans cells. In order to elucidate the mode of action of colistin in fungi we performed microarray analysis of samples treated with only aminocandin (0.00125µg/ml) or treated with aminocandin (0.00125µg/ml) and colistin (5µg/ml). We compared: (A). untreated cells to cells treated with aminocandin only; (B). cells treated with aminocandin to cells treated with aminocandin and colistin. By comparing those datasets it should be possible to identify genes differentially expressed in response to aminocandin and in response to both drugs. And subsequently to be able to interpret where in the cell colistin acts. (See related experiment in ArrayExpress: E-MEXP-3438)

Lapatinib and retinoic acid combination treatment of SKBR3 breast cancer cells

Project description:SKBR3 cells, which bear both an HER2 and a RARA gene amplification, were treated for 12 or 48 hours with 100 nM retinoic acid, 100 nM lapatinib or the combination.The two drugs synergize and induce massive apoptosis. The aim is to find the molecular mechanism(s) of this synergism. Gene expression profiling was performed using Agilent two-color 4X44K arrays. curcumin aorta apoEKO mice

Project description:At eight weeks of age, male mice were divided into two groups (15 mice per group) fed iso-energetic diet: a control-diet or the same diet supplemented with 0.2% of curcumin (Sigma Aldrich) for 16 weeks (Table S1). The dose of curcumin used in this study is equivalent to 1g curcumin consumption in humans, a level of consumption achievable in South-Asian countries. No significant difference in weight gain was observed between the two groups at the end of the experimental period (data not shown). After 16 weeks, mice were anaesthetized (40 mg pentobarbital/kg of body weight) and blood was collected from the abdominal aorta into heparinised tubes and plasma was stored at -80 C. After washing with sterilized PBS, aorta and heart samples were immediately frozen in liquid nitrogen and stored at -80 C. Project description:We generated a gene replacement allele of the E-cadherin locus that express an N-cadherin-GFP fusion in ES cells. Expression profiles of homozygous and heterozygous knock-in ES cells were analyzed in comparison to wt ES cells. The GEO Samples were compared to published data of E-cadherin knock-out ES cells (ArrayExpress, accession number E-MEXP-2836). The comparison data are linked below.

Transcriptomic analysis reveals the involvement of canonical Wnt signaling in zebrafish gonad differentiation

Project description:Zebrafish males undergo a juvenile ovary to testis type gonadal transformation process. To understand how juvenile ovary-to-testis transformation in zebrafish is genetically regulated, we compared the transcriptomes of juvenile ovary (JO) and juvenile ovotestis (JOT) samples at 35 dpf using the Gonad Uniclone Microarray v1 (ArrayExpress ID: A-MEXP-838). We also analyzed the transcriptomes of the 32 dpf JO, 32 dpf JOT, 34 dpf JOT, and 36 dpf JOT gonads using the slightly modified Gonad Uniclone Microarray v2 and together with the 35 dpf JO and 35 dpf JOT samples, compared the JO samples against the JOT samples to identify genes that could be potentially involved in the gonad transformation process.
