Transcription profiling of young and old yeast cells grown in pH controlled and uncontrolled media.

ABSTRACT: A 2 x 2 factorial design was used to elucidate the genome-wide transcriptional responses of old and young cells to the medium pH control. ?HO strain (BY4742 background) was cultivated batch-wise in SDC media and in fully controlled fermenters. pH was maintained at 4.5 via automatic NaOH addition for pH controlled case while this control was turned off for the unbuffered experiments. Samples of the young and old cells, collected at the 3rd and 7th day of the experiment respectively, were transferred to stimulating media containing fresh SDC medium prior to sample collection for transcriptional profiling.

Transcription profiling by array of an amylolytic yeast

Project description:The amylolytic S. cerevisiae strain WTPB-G and the wild-type strain (control) from a genetic background of FY23 were used to elucidate the effect brought about to a wild-type strain by the presence of an extrachoromosomal self-replicating element in the form of a recombinant multicopy plasmid. Strains were cultivated in YPD medium in fully controlled 2 L B-Braun Biostat B Plus fermenters, with a working volume of 1.5 L under aerated and micro-aerated conditions. Samples for transcriptional profiling were collected at the mid-exponential phase of growth at an OD range of 0.7-0.9, immediately frozen in liquid nitrogen and stored at -80C until RNA isolation. All experiments were carried out in duplicates.

Transcriptional remodeling in response to transfer upon carbon-limited or metformin-supplemented media in S. cerevisiae and its effect on chronological life span

Project description:The genome-wide transcriptional response of S. cerevisiae cells upon transfer to carbon-limited or metformin supplemented media is investigated. DHO strain (BY4742 background) was cultivated batch-wise in SDC media at 30C and 180 rpm up to mid-exponential phase (OD600 ~ 0.6). The cells of the main culture were divided in three aliquots at the mid-exponential phase, centrifuged at 6000 rpm for 5 min, washed twice with deionized and distilled sterile water prior to their transfer to the treatment media. Treatment media comprised of fresh SDC (2% glucose) for the control case, SDC + 0.25% glucose for the C-lim case and SDC + 1.66% metformin for the Met case. Samples collection for transcriptional profiling was done at the 2nd hour of the transfer. The experiments were carried out in biological triplicates.

Project description:Transcriptional profiles from Acidithiobacillus ferrooxidans type strain ATCC23270 grown in the presence of iron or elemental sulfur as energy source until mid logarithmic phase were compared. Cells were harvested after 48 hs of growth in 9K-FeII pH 1.6 and after 120 hs of growth in 9K-S0 pH 3.5. Pellets were washed to remove precipitates, frozen and stored at -80C until RNA extraction. In toto 8 different hybridizations were performed using 4 independent biological samples of A. ferrooxidans (with dye-swap), 4 arrays of array design FCV-CNRS AFE Oligoarray v.1 and 4 arrays of array design FCV-CNRS AFE Oligoarray v.2. This experimental design produced 8 raw data files and 1 transformed and/or normalized data file.
