Project description:To gain new insights into molecular changes in skeletal muscle aging and disease with a special focus on differential alternative splicing and senescence, we performed RNA-seq on tibialis muscles from 15 months old (adult, a) and 27 months old (old, o) rats. Whole (w) muscle samples were prepared for both age groups (w/a and w/o). Additionally, for both age groups, muscle fibers were isolated. For adult rats, this resulted in one group (f/a). For old rats, fibers were subjected to Laser capture microdissection guided by expression of aging markers Tmem158 and Cdkn1a, giving rise to two groups, aging marker-positive fibers (f/o+) and aging marker-negative fibers (f/o-). The entire experiment comprises 5 groups (w/a, w/o, f/a, f/o+, f/o-), with 5 animals per group, ie. 25 samples total.

Project description:A regular physical training is known to contribute to preserve muscle mass and strength, maintaining structure and function of neural and vascular compartments and preventing muscle insulin resistance and inflammation. However, physical activity is progressively reduced during aging causing mobility limitations and poor quality of life. Although physical exercise for rehabilitation purposes (e.g., after fractures or cardiovascular events) or simply aiming to counteract the development of sarcopenia is frequently advised by physicians, nevertheless few data are available on the targets and the global effects on the muscle organ of adapted exercise especially if started at old age. To contribute answering this question for medical translational purposes, the proteomic profile of the gastrocnemius muscle was analyzed in 24-month-old mice undergoing adapted physical training on a treadmill for 12 weeks or kept under a sedentary lifestyle condition. Proteomic data were implemented by morphological and morphometrical ultrastructural evaluations. Data demonstrate that muscles can respond to adapted physical training started at old age, positively modulating their morphology and the proteomic profile fostering protective and saving mechanisms either involving muscle cell components and pathways (i.e., mitochondrial processes, translation pathways and protein refolding, contractile apparatus) as well as the extracellular compartment. Therefore, this study provides important insights on the targets of adapted physical training, which can be regarded as suitable benchmarks for future in vivo studies further exploring the effects of this type of physical activity by functional/metabolic approaches.

Project description:In recent years, thanks to the great development of mass spectrometry (MS)-based high-throughput proteomic techniques, a large-scale protein characterization is less challenging. Proteomic approaches have been successfully applied to SM in different experimental models [21–24]; however, to date, there is little information on ECM proteins and how they change during ageing. The identification and quantification of ECM components and their interactions are es-sential steps to understand the role of the matrisome in sarcopenia. In this context, we combined a proteomic approach (i.e., LC-MS/MS and bioinformatic analyses) with mor-phological and morphometrical evaluations at fluorescence and transmission electron microscopy of the gastrocnemius muscle in adult and old mice. This age range has been selected since it has been observed that most of changes in muscle protein expression take place after middle age. The gastrocnemius muscle was selected for analysis since it is prevalently composed of fast-twitch fibre, which are especially affected by atrophy during aging. Our findings highlighted several modifications of ECM protein composition and organization in old mice, which likely play a role in the alteration of muscle properties during aging.

Project description:Few studies have investigated heterogeneity of selection response in replicate lines subjected to equivalent selection. We developed 4 replicate lines of mice based on high levels of voluntary wheel running (high runner or HR lines) while also maintaining 4 non-selected control lines. This led to the unexpected discovery of the HR mini-muscle (HRmini) phenotype, recognized by a 50% reduction in hindlimb muscle mass, which became fixed in 1 of the 4 HR selected lines. Here, we report genome-wide expression profiling describing transcriptome differences between HRnormal and HRmini medial gastrocnemius. This work provides a resource for understanding differences in muscle phenotypes in populations exhibiting high running capacity. Male mice were used from an animal model of 4 closed lines selectively bred for high voluntary wheel-running behaviour (HR) and 4 control lines bred without intentional selection, described in Swallow et al. Animals used in the current work were sampled from generation 37 at a mean age of 84 days (range = 79-86). For microarray analysis, we used gastrocnemius muscles (including both lateral and medial portions) from HRmini line #3 and HRnormal line #8 (n = 6, in each group). To obtain sufficient RNA, gastrocnemius samples of HRmini were pooled from left and right limbs from each animal, whereas HRnormal gastrocnemius was sampled from either left or right limb only.

Project description:We have previously documented a time-course of sarcopenia development in female C57Bl/6J mice, with significant loss of quadriceps muscle mass by 24 m that is more pronounced by 27-29 m. The present study used gastrocnemius muscles from female C57Bl/6J mice aged 3, 15, 24 and 29 months to investigate the expression of genes that coincide with, or potentially precede, muscle loss. The microarray analyses of ageing gastrocnemius muscle indicated that the Functional Theme Protein catabolism was first discernable in 15 and 24 age contrast and then again in the age contrast between 24 m and 29 m. A common Functional Theme in all age contrasts from the microarray analysis was extracellular matrix suggesting continual remodeling of skeletal muscle during ageing. Metabolic traits, particularly those involving altered fatty acid and glucose metabolism were apparent in the contrast between 24 m and 29 m. This may reflect inefficiency of lipid metabolism with ageing caused by altered mitochondrial function, which has been implicated in ageing processes in other studies. There were no functional themes associated with oxidative stress, inflammation or immune cell infiltration in any age contrast, likely indicating that these factors are not primary drivers of sarcopenia. Ageing was also associated with increased global gene expression variance, consistent with generalised decreased control of gene regulation. Microarray profiling was performed in gastrocnemius muscles from female C57Bl/6J mice aged 3, 15, 24 and 29 months.

Project description:This study examined the effects of castration and testosterone replacement on global differential gene transcription in the gastrocnemius muscle (m.Gas) in young adult mice over 14-days. Each of the 3 treatment groups was composed of 3 independent biological replicates, totaling 9 arrays. Equal amounts of the highest quality total RNA (4 µg) from three animals were pooled for each biological replicate, totaling the use of 27 animals in the 9-array experiment. The balanced design of this study enabled the use of a 1-way ANOVA, which was used to statistically determine which of the >34,000 genes (>45,000 probes) were differentially expressed between groups. The three pair-wise comparisons were: Castration vs. Sham (castration (C) effect); Testosterone vs. Castration (testosterone replacement (T) effect); and Testosterone vs. Sham (overall effects).
