The germ cell fate of cynomolgus monkeys is specified in the amnion

Project description:The germ cell lineage ensures reproduction, heredity and evolution. However, the mechanism for germ cell specification in primates, including humans, has been unknown. In primates, the pluripotent epiblast segregates the amnion upon implantation and thereafter initiates gastrulation to generate three germ layers. Here, we show that in cynomolgus monkeys, the SOX17/BLIMP1/TFAP2C-positive primordial germ cells (cyPGCs) arise from the dorsal amnion at embryonic day (E) 11. cyPGCs then migrate down beneath the epiblast and expand their numbers, through proliferation and continuous recruitment from the posterior amnion, around posterior yolk sac endoderm by E17. Remarkably, the amnion itself expresses BMP4, a cytokine potentially critical for PGC specification, thereby inducing cyPGCs in an autocrine fashion. Consistently, the amnion expresses T, a key mesodermal factor, prior to the onset of gastrulation. Our study demonstrates the origin of cyPGCs in the amnion, which undergoes a unique morphogenetic event prior to and independent from the gastrulation.

Epigenetic regulation and transcriptional repression by Tet1 in the early post-implantation mouse embryo: WG Bisulfite sequencing

Project description:The mammalian TET dioxygenases contribute to global waves of DNA demethylation in the zygote and in primordial germ cells, but their involvement during de novo DNA methylation at peri/post-implantation development is unknown. Here, we show novel physiological functions of Tet1 in the pre-primitive streak stage mouse embryo, where it is expressed not only in the primed-state epiblast, but also in the extra-embryonic ectoderm. In the epiblast, Tet1 contributes to DNA methylation patterning, which indirectly results in dominant transcriptional repression involving a Jumonji-family gene Jmjd8. In the extra-embryonic ectoderm, Tet1 suppresses expression of metabolic genes involved in oxidative phosphorylation. These lineage-specific gene repressive functions, involving distinct modes of regulation by DNA methylation, counteract precocious differentiation of the embryo prior to the onset of gastrulation. Such dysregulation in the absence of Tet1 are surprisingly tolerated in an inbred strain but results in full embryonic lethality in non-inbred mice, thus implicating a complex but essential role of Tet1 in normal gestational development. This dataset includes the WGBS/oxWGBS: Methylation and hydroxymethylation profiling of epiblast-like cells (EpiLCs) in presence or absence of TET1. Whole genome bisulfite sequencing and oxidative whole genome bisulfite sequencing were performed on two wt EpiLCs (male) and two TET1 knock-out EpiLCs (one male and one female).

RNA-seq data analysis of Smad5 knockout and wild type individual mouse amnion samples collected during early embryonic development

Project description:Mice deficient in the BMP-effector, Smad5 (Smad5 KO), develop severe defects in embryonic morphogenesis as well as a delay in amnion-chorion separation, important extraembryonic tissues. After closure of the proamniotic canal, a remarkable ectopic primitive streak-like aggregate develops in the amnion of these mutants. We investigated the earliest steps of mutant amnion misdifferentiation by RNAseq of single Control (Ctrl) and Smad5 KO amnion samples collected before the appearance of the aggregate. The transcriptome analysis revealed two separate sets of non-squamous amnion defects. One set of mutants (KO-SetA) robustly overexpressed streak mesoderm-related genes conform former analyses (Pereira et al., 2012). The other set overexpressed extraembryonic ectoderm markers suggestive of chorionic inclusion in amnion (KO-SetB). Tetraploid chimera analyses confirmed that SMAD5 deficiency in the epiblast can result in two distinct sets of amnion defects: one with impaired anterior amnion expansion and differentiation, and another with inclusion of chorionic extraembryonic ectoderm in the space normally occupied by amnion.

NANOG-OCT4-SOX2 Regulatory Module in Human Embryonic Stem Cells (dataset 2)

Project description:The transcription factors Nanog, Oct4 and Sox2 are the master regulators of pluripotency in mouse embryonic stem cells (mESCs), however, their functions in human ESCs (hESCs) have not been rigorously defined. Here we show that the requirements for NANOG, OCT4 and SOX2 in hESCs differ from those in mESCs. Both NANOG and OCT4 are required for self-renewal and repress differentiation. OCT4 controls both extraembryonic and epiblast-derived cell fates in a BMP4-dependent manner. OCT4-depleted hESCs commit to trophectoderm and primitive endoderm in the presence of BMP4, but undergo neuroectoderm differentiation in the absence of BMP4. NANOG represses neuroectoderm and neural crest commitment, but has little or no effect on the other lineages. We find that SOX2 is not required for self-renewal because it is redundant with SOX3, which is induced in SOX2-depleted hESCs. Simultaneous depletion of both SOX2 and SOX3 induces differentiation into the primitive streak. Unexpectedly, we identify significant variability in the usage of pluripotency factors by individual hESC lines, suggesting that the pluripotency network is remodelled to support a continuum of developmental states. Our study revises the general view of how NANOG, OCT4 and SOX2 orchestrate self-renewal in hESCs. Total RNA obtained from hESCs with or without BMP4 treatment for 8 days time course.
