Project description:Trypanosoma brucei is a kinetoplastid protozoan and the causal agent of Human African Trypanosomiasis. The life cycle is complex with multiple developmental forms. To identify gene regulations in different developmental forms, we carried out gene expression analysis of two replicative developmental forms which are readily recapitulated in laboratory culture: the ‘slender’ mammalian bloodstream form and the tsetse procyclic form.
Project description:Most protozoan pathogens of mammals are transmitted by invertebrate vectors and a comparison of transcriptomes of the developmental forms provides information on genes involved in interactions with different hosts. Trypanosoma brucei is the causal agent of African Trypanosomiasis and is transmitted by Tsetse flies. Here, cultured mammalian bloodstream forms were differentiated in vitro to insect procyclic forms and RNAseq experiments performed on the two developmental forms.
Project description:RBP9-TAP was expressed in bloodstream T. brucei and bound mRNA was pulled down using IgG beads and eluted via TEV-protease cleavage. rRNA was depleted using RNAseH and rRNA hybridizing oligos.
